摘  要:为开发高效的薏苡分子标记,加速薏苡种质资源评价和分子育种进程,本研究以贵州地方品种‘小白壳’全基因组序列为基础,在薏苡基因组序列上识别出7 872 192个SSR位点,平均0.2 kb出现一个SSR位点。其中,单核苷酸为主要重复类型,占总SSR的98.42%,其次为三核苷酸和二核苷酸,分别占总SSR的0.76%和0.74%。6种核苷酸基元的重复次数主要在5~6次,所占比例分别为74.27%和15.17%。以三核苷酸重复基元为对象,随机选择均匀分布在薏苡基因组的200个SSR位点,利用毛细管电泳技术在9个不同类型薏苡材料中对SSR标记的有效性进行检测。结果表明,173对引物有扩增产物,47对引物能够获得清晰的目的条带且具有多态性,有效引物率为23.5%。每对引物平均等位基因数为3.3,PIC值为0.20~0.84,平均为0.60。本研究结果表明薏苡全基因组SSR标记数量丰富,类型多样,开发的具有多态性的标记为薏苡种质资源鉴定、亲缘关系分析和分子标记辅助育种提供了有力工具。In or der to develop efficient molecular markers to accelerate germplasm evaluation and molecular breeding in Coix lacryma-jobi L., a total of 7 872 192 SSR loci were identified from the genome sequence of Coix lacryma-jobi L., a local variety in Guizhou Province, with an average of 0.2 kb. Mononucleotide was the main repeat type, accounting for 98.42% of the total SSR, followed by trinucleotide and dinucleotide, accounting for0.76% and 0.74% of the total SSR, respectively. The repeats of the 1~6 nucleotide units were mainly 5~6 times, accounting for 74.27% and 15.17%, respectively. Taking trinucleotide repeat units as the object, 200 SSR sites evenly distributed through the genome were randomly selected and detected in 9 different types of Coix lacryma-jobi L. by capillary electrophoresis. The results showed that 173 pairs of primers had amplification products, of which 47 pairs of primers could obtain clear target bands with polymorphism, and the effective primer rate was 23.5%. The average number of alleles per primer pair was 3.3, and the PIC value ranged from 0.20 to 0.84 with an average of0.60. The results of t his study showed that the SSR markers in the whole genome of Coix lacryma-jobi L. were abundant and varied in type. The polymorphic markers developed provided a powerful tool for germplasm identifi cation, genetic relationship analysis and molecular marker-assisted breeding.
