摘  要:为了研究摄入不同水平的呕吐毒素(DON)对小鼠血清及不同小肠段免疫水平和抗氧化水平的影响,试验将80只SPF昆明系小鼠随机分为4组,每组20只,低、中、高剂量组分别灌胃浓度为1.0,1.5,2.5 mg/kg的DON,对照组灌胃等量的生理盐水,连续灌胃21 d。试验结束后计算平均日增重、平均日采食量、料重比,采集血清及十二指肠、空肠和回肠肠段,采用ELISA法测定免疫球蛋白[免疫球蛋白(Ig)A、IgG、分泌型免疫球蛋白(sIg)A)]和抗氧化指标[总抗氧化能力(T-AOC)、谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、丙二醛(MDA)]。结果表明:高剂量组的平均日增重、平均日采食量均极显著低于对照组(P<0.01),而各组之间料重比差异不显著(P>0.05)。与对照组相比,高剂量组血清中IgA含量极显著升高(P<0.01),IgG含量显著升高(P<0.05),十二指肠sIgA含量显著降低(P<0.05),回肠sIgA含量极显著降低(P<0.01)。与对照组相比,高剂量组血清中T-AOC极显著降低(P<0.01),SOD活性显著降低(P<0.05),高剂量组十二指肠、空肠、回肠T-AOC、GSH-Px活性显著降低(P<0.05);低、中、高剂量组回肠SOD活性均显著降低(P<0.05),高剂量组十二指肠和空肠的SOD活性显著降低(P<0.05);低、中、高剂量组十二指肠MDA含量显著升高(P<0.05),高剂量组空肠MDA含量显著升高(P<0.05)。说明随着灌胃DON浓度的升高,十二指肠和回肠的黏膜免疫功能显著降低,对小肠的抗氧化能力影响较大,十二指肠最易受到影响;中、高剂量的DON激活了机体的免疫功能;DON降低了机体的抗氧化能力,但并不能引起机体明显的抗氧化水平变化。To study the effects of different levels of deoxynivalenol(DON) on immune levels and antioxidant levels in mice serum and different intestinal, 80 SPF Kunming mice were randomly divided into 4 groups, with 20 mice in each group. The animals in low, medium, and high-dose groups were given intragastrically 1.0, 1.5, and 2.5 mg/kg DON, and those in the control group were given the same amount of normal saline for 21 days. After the test, the average daily gain, the average daily feed intake and the feed-to-weight ratio were calculated. Serum, duodenum, jejunum and ileum tissue were collected, and the levels of IgA, IgG, sIgA and antioxidant index(total antioxidant capacity [T-AOC], glutathione peroxidase [GSH-Px], superoxide dismutase [SOD], malondialdehyde [MDA]) were determined by enzyme-linked immunosorbent assay. The results showed that the average daily gain and average daily feed intake in the high-dose group were significantly lower than those in the control group(P<0.01), while the feed-to-weight ratio between the groups was not significantly different(P>0.05). Compared with the control group, the serum IgA content in the high-dose group increased significantly(P<0.01), the IgG content increased significantly(P<0.05), and the duodenal sIgA content decreased significantly(P<0.05). The ileum sIgA content reduced extremely significantly(P<0.01). Compared with the control group, the serum T-AOC activity in the high-dose group reduced significantly(P<0.01), so was the level of SOD activity(P<0.05). The activities of T-AOC and GSH-Px in duodenum, jejunum and ileum in high-dose group decreased significantly(P<0.05). The SOD activity of ileum in the low, medium, and high-dose groups reduced significantly(P<0.05), and the SOD activity in the duodenum and jejunum in the high-dose group reduced significantly(P<0.05). The MDA content in the duodenum in the low, medium and high dose groups increased significantly(P<0.05), and the MDA content in the jejunum in the high dose group increased significantly(P<0.05).
