摘  要:为研究特克赛尔×哈萨克横交F2代绵羊Myf5基因多态性(single nucleotide polymorphisms, SNPs)及其与胴体性状之间的关系,试验采用PCR测序技术和飞行时间质谱检测分型方法对434只特克赛尔×哈萨克横交F2代绵羊的Myf5基因SNPs进行分型,并将分型结果与其胴体性状(胴体重、胴体长、踝骨宽、臀宽、胸宽、肩宽和胸深)进行关联分析。结果表明:在Myf5基因上共发现了5个SNPs位点,即A116460176G、rs604841316、rs416158998、rs588855095和rs412277497。其中rs416158998属于内含子突变,在特克赛尔×哈萨克横交F2代绵羊中表现为中度多态[0.25<多态信息含量(PIC)<0.5],不处于Hard-Weinberg平衡状态(P≤0.05),与胸宽、肩宽和臀宽呈显著或者极显著相关(P<0.05或P<0.01)。rs588855095属于内含子突变,在绵羊中表现为低度多态(PIC≤0.25),处于Hard-Weinberg平衡状态(P>0.05),与臀宽呈显著相关(P<0.05)。rs412277497属于外显子上丝氨酸到半胱氨酸的错义突变,在绵羊中表现为中度多态(0.250.05),与胴体长、胸宽和肩宽呈显著或极显著相关(P<0.05或P<0.01)。A116460176G和rs604841316与胴体性状相关性不显著(P>0.05)。说明Myf5基因中rs416158998、rs588855095和rs412277497位点可以为培育优良新品种绵羊提供可参考的遗传标记。In order to study the rlationship between singe nucleotide polymorphisms(SNP's)of My(5 gene and careas tnits of Texelx Kazakh Cross F2 generation sheep,PCR sequencing technology and time-of-flight mass spectrometry dection method were used to type Myf5 gene SNPs in 434 Texelx Kazakh cross F2 generation sheeps,and correlation analysis was conducted between the easifieation resuls and is careass trails(carcass weight,careass length,ankle width,hip width,chest width,shoulder width and chest depth).Resuls showed that a total of five.SNPs loci were found in the Myf5 gene,namely Al 16460176C,r>604841316.rs416158998.r588855095 and rs412277497.Among them,r>416158998,an intron mutation,was moderately polymorphice in Texelx Kazakh cross F2 generation sheep(0.25<PIC<0.5).not in a Hard-W einberg equilibrium state(P≤0.05),and signifcantly or extremelly signifcanly orrelated with chest width,shoulder width and hip width(P<0.05 or P<0.01).r588855095,an intron mutation,was low polymophism in sheep(PIC≤0.25),in a Hard-Weinberg equilibrium state(P>0.05),and signifcantly correlated with hip widh(P<0.05).r412277497.a missense mutation with serine mutated to cystein on the exon,was moderately polymorphie in sheep(0.25<PIC<0.5).in a Hard-W einberg equilibriumn state(P>0.05).and significantly or ex-tremelly signifcantly crrelated with carcass lengh,chest width and shoulder widh(P<0.05 or P<0.01).A1164601766和rs604841316 was not signifieantly correlated with earcass trais(P>0.05).The experiment revealed that n416158998,r588855095 and rs412277497 can provide reference genetic markers for breeding new breeds of sheep.
