摘  要:目的研究环状非编码RNA基因PVT1(circPVT1)在鼻咽癌组织中的表达及对鼻咽癌细胞增殖、转移和凋亡的影响,并探讨其作用机制。方法采用实时荧光定量PCR(RT-qRCR)检测circPVT1在鼻咽癌组织和鼻窦炎组织(对照组)中的表达,将鼻咽癌细胞系HNE1体外培养并分成阴性对照组(转染阴性对照载体)、circPVT1过表达试验组(转染circPVT1 mimic)和circPVT1干扰试验组(转染circPVT1 inhibitor)。采用RT-qRCR检测circPVT1 mimic和circPVT1 inhibitor转染效果;采用MTT试验分析circPVT1对鼻咽癌细胞增殖的影响;采用Transwell chamber试验分析circPVT1对鼻咽癌细胞转移的影响;流式细胞术分析circPVT1对鼻咽癌细胞凋亡的影响;采用Western blot分析circPVT1对PTEN-PI3K/AKT信号通路表达的影响。结果与对照组相比,鼻咽癌组织中circPVT1表达明显上调(P<0.05)。转染circPVT1 mimic后,circPVT1过表达试验组细胞中circPVT1的表达增加(P<0.05),转染circPVT1 inhibitor后,circPVT1干扰试验组细胞中circPVT1的表达降低(P<0.05)。转染circPVT1 mimic显著促进鼻咽癌细胞的增殖和转移能力,抑制细胞凋亡,同时降低PTEN表达和增加PI3K/AKT磷酸化(P<0.05)。转染circPVT1 inhibitor显著抑制鼻咽癌细胞的增殖和转移能力,促进细胞凋亡,同时增加PTEN的表达和降低PI3K/AKT磷酸化(P<0.05)。结论circPVT1在鼻咽癌中表达增加,可通过靶向调节PTEN-PI3K/AKT信号途径促进鼻咽癌细胞增殖和转移能力,并抑制细胞凋亡,进而参与鼻咽癌的发展,circPVT1可能是治疗鼻咽癌的潜在新靶点。Objective To study the expression of circular non-coding RNA gene PVT1(circPVT1)in nasopharyngeal carcinoma tissues and its effect on the proliferation,metastasis and apoptosis of nasopharyngeal carcinoma cells,and to explore its mechanism of action.Methods Real-time fluorescence quantitative PCR(RT-qRCR)was used to detect the expression of circPVT1 in nasopharyngeal carcinoma tissues and sinusitis tissue(control group).The nasopharyngeal carcinoma cell line HNE1 was cultured in vitro and divided into control groups(transfected with negative control vector),circPVT1 overexpression experimental group(transfected with circPVT1 mimics)and circPVT1 interference experimental group(transfected with circPVT1 inhibitor).RT-qRCR was used to detect the transfection effect of circPVT1 mimics and circPVT1 inhibitor.MTT experiment was used to analyze the effect of circPVT1 on the proliferation of nasopharyngeal carcinoma cells.Transwell chamber experiment was used to analyze the effect of circPVT1 on the metastasis of nasopharyngeal carcinoma cells.Flow cytometry was used to analyze the effect on the apoptosis of circPVT1 on nasopharyngeal carcinoma.The influence of circPVT1 on the expression of PTEN-PI3K/AKT signal pathway was analyzed by Western blot.Results Compared with control group,the expression of circPVT1 in nasopharyngeal carcinoma tissues was significantly up-regulated(P<0.05).After transfection of circPVT1 mimic,the expression of circPVT1 in the cells of the circPVT1 overexpression experimental group were increased(P<0.05).After transfection of circPVT1 inhibitor,the expression of circPVT1 in the cells of the circPVT1 interference experimental group were decreased(P<0.05).Transfection of circPVT1 mimic significantly promoted the proliferation and metastasis of nasopharyngeal carcinoma cells,inhibited cell apoptosis,reduced PTEN expression and increased PI3K/AKT phosphorylation(P<0.05).Transfection with circPVT1 inhibitor significantly inhibited the proliferation and metastasis of nasopharyngeal carcinoma cells,pro
