摘 要:目的评价益肾骨康方对骨转移癌痛大鼠的骨保护作用。方法48只SD大鼠随机分为对照组、模型组、解毒化瘀组、扶正组、益肾骨康方低剂量组、益肾骨康方高剂量组,每组8只。除对照组外其余各组采用大鼠乳腺癌MRMT-1细胞悬液10μL缓慢注射于大鼠胫骨骨髓腔内建立骨转移癌模型。于造模后第2天开始给药,中药组给予相应中药1 mL/100 g灌胃,对照组及模型组给予等量生理盐水灌胃,每日1次,连续2周。观察各组大鼠体质量变化,胫骨HE染色观察骨质破坏情况。结果给药2周后,与模型组比较,扶正组及低剂量组体质量下降不明显(P<0.05);从胫骨病理组织学上可以看出,模型组骨组织被肿瘤细胞破坏严重,解毒组及扶正组骨髓腔内可见少量肿瘤细胞浸润,骨小梁及骨皮质受损程度较模型组减轻。低剂量组及高剂量组骨皮质较完整,髓腔内可见肿瘤细胞,骨小梁受损较轻。结论益肾骨康方可减轻骨转移癌导致的溶骨性骨破坏。Objective To evaluate the bone protection eff ect of Yishen Gukang prescription on rats with the bone metastasis cancer pain.Methods A total of forty-eight SD rats were randomly divided into a control group,a model group,a Jiedu Huayu group,a Fuzheng group,a low-dose Yishen Gukang group and a high-dose Yishen Gukang group,with 8 rats in each group.Except the control group,10μL of the rat breast cancer MRMT-1 cell suspension was slowly injected into the bone marrow cavity of the rat tibia to establish the model of bone metastases.On the second day after modeling,the groups treated with Chinese materia medicas were given 1 mL/100 g of corresponding traditional Chinese drugs by gavage,while the control group and the model group were given the same amount of normal saline by gavage,once a day,lasting for consecutive two weeks.The changes of body mass in each group were observed,and the bone destruction was observed by tibia HE staining.Results After two weeks of administration,compared with the model group,the body weight of the Fuzheng group and the low-dose group was not signifi cantly decreased(P<0.05).According to the histomathology of tibia,the bone tissue of the model group was severely damaged by tumor cells,while a small number of tumor cells were found in the bone marrow cavity of the Jiedu group and the Fuzheng group,and the damage to the bone trabecula and bone cortex was less severe than that of the model group.In the low-dose and high-dose groups,the bone cortex was intact,while the tumor cells were found in the medullary cavity,and the bone trabecula was less damaged.Conclusion Yishen Gukang prescription can alleviate the osteolytic destruction caused by bone metastases.
