摘 要:目的探索短期高浓度葡萄糖摄入对小鼠实验性自身免疫性葡萄膜炎(EAU)发展的促进作用。方法选取健康SPF级6~8周龄小鼠38只,随机分为模型对照组(EAU组)和高糖模型组(GLU组),每组19只小鼠。GLU组小鼠从造模前14 d开始给予200 g·L^(-1)葡萄糖溶液饮水直至处死,EAU组小鼠给予正常饮水,同时保证EAU组和GLU组小鼠每日摄入饲料量相同。造模后14 d,用裂隙灯对各组小鼠进行临床评分;分离小鼠眼球组织,HE染色进行各组小鼠组织病理学评分;分离小鼠视网膜组织,QT-PCR检测各组小鼠视网膜IL^(-1)7和IFN-γ的相对表达量;分离小鼠眼球和血清,ELISA检测各组小鼠眼球和血清中IL^(-1)7和IFN-γ的蛋白水平;分离脾脏,流式细胞仪检测各组小鼠脾脏中Th17细胞占CD4+细胞的比例。结果造模后14 d,GLU组小鼠临床评分为(3.00±0.27)分,高于EAU组[(1.58±0.25)分],差异有统计学意义(P=0.002);GLU组小鼠组织病理学评分为(3.08±0.23)分,高于EAU组[(1.88±0.30)分],差异有统计学意义(P=0.008)。QT-PCR结果显示,GLU组小鼠视网膜IL^(-1)7和IFN-γ的相对表达量均高于EAU组,IL^(-1)7表达差异有统计学意义(P=0.017),IFN-γ表达差异无统计学意义(P=0.081)。ELISA结果显示,GLU组小鼠眼球和血清IL^(-1)7含量分别为(102.70±8.32)ng·L^(-1)、(113.20±6.80)ng·L^(-1),均高于EAU组[(78.23±5.79)ng·L^(-1)、(82.34±2.48)ng·L^(-1)],差异均有统计学意义(P=0.030、0.002),而两组小鼠眼球和血清中IFN-γ水平差异均无统计学意义(P=0.781、0.589)。流式细胞仪检测结果显示,GLU组小鼠脾脏Th17细胞占CD4+细胞比例为(2.15±0.06)%,高于EAU组[(1.81±0.06)%],差异有统计学意义(P=0.007)。结论短期高浓度葡萄糖摄入可以加重EAU小鼠模型的炎症,增加眼内IL^(-1)7的表达,其可能与外周血中Th17细胞的过度激活有关。Objective To explore the promoting effect of short-term high glucose intake on the development of experimental autoimmune uveitis(EAU)in mice.Methods Thirty-eight healthy SPF mice aged 6 to 8 weeks were randomly divided into the model control group(EAU group)and high glucose model group(GLU group),with 19 mice in each group.Mice in the GLU group were given 200 g·L^(-1) glucose solution from 14 days before modeling until death,while mice in the EAU group were given normal water.Mice in both groups were fed the same daily.On day 14 after modeling,the mice in each group were scored with the slit lamp.Eyeball tissues were separated,and their histopathological score was obtained by hematoxylin-eosin staining.Retinal tissues were isolated,and the relative expression levels of interleukin-17(IL^(-1)7)and interferon-γ(IFN-γ)in the retina were detected by quantitative polymerase chain reaction(QT-PCR).Eyeballs and serum were separated,and the protein levels of IL^(-1)7 and IFN-γin the eyeballs and serum were measured by ELISA.The spleen was isolated,and the ratio of T helper(Th)17 cells to CD4+cells was calculated by flow cytometry.Results On day 14 after modeling,the clinical score of mice in the GLU group was 3.00±0.27,which was higher than that in the EAU group(1.58±0.25),and the difference was statistically significant(P=0.002).The histopathological score of mice in the GLU group was 3.08±0.23,which was higher than that in the EAU group(1.88±0.30),and the difference was statistically significant(P=0.008).QT-PCR results showed that the relative expression levels of IL^(-1)7 and IFN-γin the retina of mice in the GLU group were higher than those in the EAU group.The difference in IL^(-1)7 between the two groups was statistically significant(P=0.017),but the difference in IFN-γwas not statistically significant(P=0.081).ELISA results showed that the IL^(-1)7 content in eyeballs and serum of mice in the GLU group were(102.70±8.32)ng·L^(-1) and(113.20±6.80)ng·L^(-1),respectively,which were both higher than thos
