摘　　要：目的探究体外LINC00847表达对乳腺癌细胞增殖、迁移和侵袭的影响。方法应用shRNA干扰乳腺癌细胞株MCF-7和MDA-MB-231中LINC00847的表达后,qRT-PCR法检测shRNA的干扰效率。确认干扰效率后分别使用MTT法、克隆形成实验和Transwell小室实验检测干扰LINC00847后MCF-7和MDA-MB-231细胞的增殖能力、克隆形成能力、迁移以及侵袭能力的改变。结果在MCF-7和MDA-MB-231细胞中,与对照组相比,稳染LINC00847-shRNA后LINC00847的表达明显降低(P<0.001)。与对照组相比,敲低LINC00847表达可显著下调乳腺癌细胞MCF-7和MDA-MB-231的增殖、迁移以及侵袭能力(P<0.001)。结论沉默LINC00847的表达可抑制乳腺癌的增殖、迁移以及侵袭能力。LINC00847表达可能参与了乳腺癌的发生、发展。Purpose To explore the effect of LINC00847 expression on the proliferation,migration and invasion of breast cancer cells in vitro.Methods shRNA was used to interfere the expression of LINC00847 in breast cancer cell line MCF-7.qRT-PCR was used to detect whether shRNA can interfere with the expression of LINC00847.MTT assay,clone formation experiment,and transwell assay were used to detect the changes of MCF-7 cell proliferation ability,clone formation ability,migration and invasion ability after interference with the expression of LINC00847.Results LINC00847-shRNA significantly interfered with LINC00847 expression in breast cancer cells MCF-7(P<0.001).Compared with the control group,the interference of LINC00847 expression significantly decreased the proliferation,migration and invasion capacity of MCF-7 in breast cancer cells(P<0.001).Conclusion LINC00847 expression may be involved in the occurrence and development of breast cancer.
