摘 要:目的:研究大米草黄酮(Spartina anglica flavone,SAF)的体外抗菌效果及机理。方法:以铜绿假单胞菌等5种常见临床病原菌为试验菌株,采用牛津杯法测量抑菌圈并观察SAF的药物敏感性;采用微量稀释法测定SAF的最小抑菌浓度(MIC)及最小杀菌浓度(MBC);测量经5 g/L SAF处理后的铜绿假单胞菌菌液中的磷含量,并通过SDS-PAGE分析铜绿假单胞菌蛋白质的表达情况,初步探讨了SAF的抑菌机理。结果:SAF对铜绿假单胞菌具有一定的抑制作用,对其余4种细菌无明显抑制作用;经SAF处理后,细菌对磷的消耗量降低,通过SDS-PAGE分析,证实SAF可能影响铜绿假单胞菌蛋白质的正常表达。结论:SAF对铜绿假单胞菌有一定的抗菌活性,其抑菌机制可能与SAF对细菌细胞膜的破坏力相关。Objective:In order to study the antibacterial effect in vitro and mechanism of Spartina anglica flavone(SAF).Methods:Use five common clinical pathogenic bacteria,such as Pseudomonas aeruginosa was selected as the test strains to observe the drug sensitivity of SAF by oxpord cup method.Determination of MIC and MBC of SAF was applied in determining and monitoring by the micro-dilution method,anthrone method.The content of phosphorus in Pseudomonas aeruginosa solution treated with 5 g/L SAF and the protein expression of Pseudomonas aeruginosa were measured by SDS-PAGE,and the bacteriostatic mechanism of SAF was discussed.Results:SAF showed antibacterial effect on Pseudomonas aeruginosa,while it has no significant effect on other strains.And total polysaccharide concentration of the sensitive bacterrial liquids gradually increase with the extending of antibacterial effect time,the results of SDS-PAGE showed that SAF might affect the normal expression of Pseudomonas aeruginosa protein.Conclusion:SAF shows antibacterial effect on Pseudomonas aeruginosa,the mechanism might be related to its destruction on bacterial membranes.
