摘 要:目的:探讨钆塞酸增强MRI(EOB-MRI)肝癌成像中的转运蛋白有机阴离子转运多肽IB3(OATP1B3)表达水平,并分析其与肝细胞癌(HCC)临床亚类之间的关系。方法:选取2016年4月至2017年12月接受手术治疗的HCC患者为研究对象,通过定量实时逆转录聚合酶链反应(qRT-PCR)和免疫组织化学检测临床样本中转运蛋白、转录因子和Wnt靶基因的表达水平。在体外对HCC细胞系KYN-2进行氯化锂(LiCl)处理,以评估其对Wnt信号转导活性的影响。结果:对转运蛋白mRNA和蛋白表达的分析表明,在EOB-MRI的肝胆期图像中,OATP1B3与肿瘤增强呈相关性。关联分析显示OATP1B3的表达与Wnt/β-catenin靶基因的表达呈显著相关。此外,LiCl处理诱导了KYN-2细胞中OATP1B3 mRNA的表达,在EOB-MRI中肿瘤增强预测Wnt/β-catenin活化的HCC的灵敏度和特异性分别为79.0%和82.1%。结论:在EOB-MRI中,OATP1B3是介导HCC增强的重要转运蛋白。OATP1B3的表达与Wnt/β-catenin靶基因的表达密切相关,因此HCC中OATP1B3上调很可能代表Wnt/β-catenin激活HCC的特定亚类。Objective:To investigate the expression of OATP1 B3 in liver cancer imaging by gadoxetic acid-enhanced MRI(EOB-MRI)and to stratify the molecular subclass of HCC.Methods:HCC patients who received surgical treatment from April 2016 to December 2017 were selected as study subjects.The expression levels of transporters,transcription factors,and Wnt signal pathway target genes in clinical samples were examined by quantitative real-time reverse transcription polymerase chain reaction(qRT-PCR) and immunohistochemistry.Results:The analysis of mRNA and protein expression of transporter showed that in hepatobiliary images of EOB-MRI,the expression of OATP1 B3 was associated with tumor enhancement.Association analysis of OATP1 B3 expression showed a significant correlation with the expression of Wnt/β-catenin pathway target gene.In addition,LiCl treatment induced KYN-2 cells,indicating that OATP1 B3 expression was strongly associated with the Wnt/β-catenin signal pathway.In addition,the sensitivity and specificity of using tumor enhancement to predict activation of Wnt/β-catenin for HCC in EOB-MRI were 79.0 % and 82.1%,respectively.Conclusion:In EOB-MRI,OATP1 B3 was the important transporter involved in mediating HCC enhancement.OATP1 B3 expression was closely related to the expression of Wnt/β-catenin target genes,OATP1 B3 up-regulated HCC likely represents a specific subclass of Wnt/β-catenin activated HCC.
