摘 要:目的建立一种同时检测油脂类药用辅料中胆甾醇、豆甾醇和β-谷甾醇含量的高效液相色谱法。方法采用Agilent Inspire PHP(250 mm×4.6 mm,5μm)色谱柱,流动相是甲醇和水(90∶10,V/V),等度洗脱,流速1.0 mL·min^(-1),检测波长210 nm,色谱柱温度30℃。结果3种甾醇组分在质量浓度范围1~20μg·m^(L-1)具有良好的线性范围(R^(2)>0.9999)。胆甾醇、豆甾醇和β-谷甾醇的加样回收率分别在96.02%~97.58%、96.70%~99.47%、97.34%~100.2%,RSD分别是1.7%、1.4%、1.4%。结论该方法灵敏度高,简便快速且稳定性好,线性范围宽,分析成本低,适合含有上述3种甾醇组分的药用辅料、中药、药品或食品中定性和定量分析。Objective To simultaneously determine cholesterol,stigmasterol andβ-sitosterol in oil pharmaceutical excipients by HPLC.Methods The chromatographic procedure was carried out on the Agilent Inspire PHP(250 mm×4.6 mm,5μm)column.The mobile phase consisted of methanol and water(90∶10,V/V)at a flow of 1.0 mL·min^(-1).The detection wavelength was 210 nm.The column temperature was 30℃.Results The three phytosterols components showed good linearitity at 1~20μg·m^(L-1)(R^(2)>0.9999).The recovery of cholesterol,stigmasterol,andβ-sitosterol was 96.02%~97.58%,96.70%~99.47%,and 97.34%~100.2%.The RSD was 1.7%,1.4%,and 1.4%,respectively.Conclusion This method is stable,sensitive,and convenient,with low cost and wide linear range.It is suitable for the qualitative and quantitative analysis of pharmaceutical excipients,traditional Chinese medicine,drug or food containing the above three phytosterols.
