摘  要:目的探讨miR-186过表达对肺腺癌细胞生长的影响及其是否与调节Shp2和PI3K/Akt/mTOR信号通路有关。方法将人肺腺癌细胞A549分为Con-mimic组、miR-186-mimic组和Con组,采用CCK8细胞增殖实验检测miR-186对细胞增殖的影响;采用划痕实验检测miR-186对细胞转移的影响;采用Transwell细胞侵袭实验检测miR-186对细胞侵袭能力的影响;采用RT-PCR和Western Blot检测miR-186对Shp2基因mRNA和蛋白水平的影响;采用Western blot检测PI3K/Akt/mTOR信号通路相关蛋白表达水平。结果相较Con-mimic组和Con组,miR-186-mimic组的细胞集落形成率即增殖率均极显著下调(P<0.01);相较Con-mimic组和Con组,miR-186-mimic组的细胞转移能力均极显著受限(P<0.01);相较Con-mimic组和Con组,miR-186-mimic组的细胞侵袭能力均极显著受限(P<0.01)。相较Con-mimic组和Con组,miR-186-mimic组的Shp2的mRNA和蛋白表达水平均极显著下调(P<0.01);相较Con-mimic组和Con组,miR-186-mimic组的PI3K、p-AKT和p-mTOR蛋白表达水平均显著下调(P<0.05)。结论miR-186可以通过下调Shp2基因表达和激活PI3K/Akt/mTOR信号通路从而实现对肺腺癌细胞增殖、侵袭和迁移的抑制作用。Objective To investigate the effect of Mir-186 overexpression on the growth of lung adenocarcinoma cells and whether it is related to the regulation of Shp2 and PI3K/Akt/mTOR signaling pathways.Methods Human lung adenocarcinoma A549 cells were divided into Con-mimic group,miR-186-mimic group and Con group.The effect of miR-186 on cell proliferation was detected by CCK8 cell proliferation assay.The miR-186 cells were detected by scratch test.The effect of metastasis was detected by Transwell cell invasion assay.The effect of miR-186 on cell invasion ability was detected by RT-PCR and Western Blot.The effect of miR-186 on the mRNA and protein levels of Shp2 gene was detected by Western blot.PI3K/Akt/mTOR signaling pathway-related protein expression levels was detected by Western blot.Results Compared with Con-mimic group and Con group,the cell colony formation rate and proliferation rate of miR-186-mimic group were significantly down-regulated(P<0.01);compared with Con-mimic group and Con group,miR-186-mimic The cell transfer ability of the group was significantly restricted(P<0.01).Compared with the Con-mimic group and Con group,the cell invasion ability of the miR-186-mimic group was significantly restricted(P<0.01);In the mimic group and the Con group,the mRNA and protein expression levels of Shp2 in the miR-186-mimic group were significantly down-regulated(P<0.01);compared with the Con-mimic group and the Con group,the PI3K of the miR-186-mimic group,The expression levels of p-AKT and p-mTOR protein were significantly down-regulated(P<0.05).Conclusion miR-186 can inhibit the proliferation,invasion and migration of lung adenocarcinoma cells by down-regulating the expression of Shp2 gene and activating the PI3K/Akt/mTOR signaling pathway.
