摘  要:[目的]本试验旨在探究日粮中添加富含多不饱和脂肪酸的15%紫苏籽(PFS)对湖羊肝脏自噬和凋亡的影响。[方法]选取30只体重相近且健康状况良好的2月龄湖羊公羔,随机分为2组,分别饲喂含0%(对照)和15%PFS的日粮。饲养84 d后,每组随机选择5只羊屠宰,采集肝脏组织。通过qPCR和Western blot等技术检测肝脏中自噬、凋亡及LKB1-AMPK-MTOR信号通路相关基因与蛋白的表达水平。[结果]与对照组相比,15%PFS显著上调PPARGC1A、BECN1、ATG5、ATG7基因和PPARGC1A、BECN1蛋白表达水平(P<0.05),对凋亡相关基因的表达水平无显著影响(P>0.05);15%PFS组肝脏组织中LKB1-AMPK-MTOR通路关键基因LKB1、AMPK和MTOR mRNA表达水平显著高于对照组(P<0.05)。[结论]紫苏籽可能通过调控LKB1-AMPK-MTOR信号通路和自噬相关基因与蛋白的表达水平,进而调控湖羊肝脏自噬进程。[Objectives]The experiment was conducted to investigate the effects of perilla frutescens seed(PFS)supplementation on the expression levels of genes and proteins related to autophagy and apoptosis in liver of Hu sheep.[Methods]Thirty lambs(about 2 month-old and 23 kg live weight)were randomly selected and divided into 2 groups:control group(0%FPS)and 15%PFS group.On day 84 of the experiment,five lambs in each group were randomly selected and slaughtered,and liver tissue samples were collected to analyze the expression levels of genes and proteins related to autophagy,apoptosis and LKB1-AMPK-MTOR pathway by qPCR and Western blot.[Results]Compared with control group,15%PFS significantly up-regulated autophagy-related gene PPARGC1A,and increased BECN1,ATG5 and ATG7 mRNA,and PPARGC1A,BECN1 protein expression levels(P<0.05),15%PFS had no valuable contribution to apoptosis-related genes(P>0.05).Furthermore,15%PFS altered the expression levels of key genes involved in LKB1-AMPK-MTOR pathway,including up-regulated LKB1,AMPK and MTOR mRNA expression levels when compared with control group(P<0.05).[Conclusions]PFS may regulate autophagy process in liver by altering gene and protein expression levels involved in LKB1-AMPK-MTOR pathway and autophagy.
