摘 要:目的探讨三七叶苷(PPD-25-OH)对糖尿病肾病(DNP)大鼠肾功能的改善和对炎性趋化因子的影响,以初步阐明PPD-25-OH对DNP保护作用的可能机制。方法采用链脲佐菌素诱导构建大鼠DNP模型,分为DNP组(生理盐水)、PPD低剂量组(PPD-L组,10 mg/kg PPD-25-OH)、PPD中剂量组(PPD-M组,30 mg/kg PPD-25-OH)及PPD高剂量组(PPD-H组,50 mg/kg PPD-25-OH),每组10只,另选取10只SD大鼠作为空白阴性对照组(NG组,生理盐水),进一步测定PPD-25-OH对大鼠一般状态、肾功能、肾脏病理形态的保护作用。采用ELISA测定炎性细胞因子水平变化,包括基质细胞衍生因子-1(SDF-1)、环氧化酶-2(COX-2);Western blot观察PPD-25-OH对单核细胞趋化因子(MCP-1)及结缔组织生长因子(CTGF)蛋白表达影响,初步探讨PPD-25-OH对DNP大鼠肾脏保护作用的可能机制。结果不同剂量PPD-25-OH均能降低DNP模型大鼠血肌酐、尿素氮、尿蛋白和肾脏指数水平,与DNP组比较,差异有统计学意义(P<0.05)。与DNP组比较,PPD-L、M、H组SDF-1、COX-2及MCP-1、CTGF蛋白表达水平更低,差异有统计学意义(P<0.05)。结论PPD-25-OH通过下调炎性趋化因子表达实现对DNP的保护作用。Objective To investigate the effect of panax notoginseng saponins(PPD-25-OH)on renal function and inflammatory chemokines in Diabetic nephropathy(DNP)rats,and to elucidate the possible mechanism of the protective effect of PPD-25-OH on DNP.Methods The DNP rat model was induced by streptozotocin.The rats were divided into the DNP group(normal saline),the PPD low dose group(PPD-L group,10 mg/kg PPD-25-OH),the PPD medium-dose group(PPD-M group,30 mg/kg PPD-25-OH),the PPD high dose group(PPD-H group,50 mg/kg PPD-25-OH)with 10 rats in each group,and another 10 SD rats were selected as the blank negative control group(NG group,normal saline).The protective effects of different doses of PPD-25-OH on the general state,renal function and renal pathological morphology of rats were further determined.In addition,ELISA was used to determine the changes of inflammatory cytokines,including the contents of stromal cell-derived factor 1(SDF-1)and cyclooxygenase-2(COX-2).The effects of PPD-25-OH on monocyte chemotactic factor(MCP-1)and connective tissue growth factor(CTGF)were studied by Western blotting.So as to explore the possible mechanism of the protective effect of PPD-25-OH on the diabetic kidney.Results Different doses of PPD-25-OH could decrease serum creatinine,blood urea nitrogen,urinary protein and renal index,of the DNP model,which was significantly different from those in the DNP group(P<0.05).Compared with the DNP group,the SDF-1,COX-2,MCP-1 and protein expression levels of CTGF in the PPD-M group,the PPD-L and the PPD-H group were significantly lower(P<0.05).Conclusion PPD-25-OH can protect DNP by down-regulate the expression of inflammatory chemokines.
