摘  要:采用复合蛋白酶(protamex)和碱性蛋白酶(alcalase)协同水解玉米谷蛋白,制备玉米谷蛋白水解物(corn gluten hydrolysate,CGH),研究了玉米谷蛋白水解物的体外抗氧化活性,利用乙醇诱导LO2细胞氧化损伤模型对玉米谷蛋白水解物的抗氧化应激作用进行研究。结果表明,玉米谷蛋白水解物具有良好的体外抗氧化活性,其羟自由基(·OH)清除率和DPPH自由基清除率和Fe^(2+)螯合能力的半抑制浓度(half maximal inhibitory concentration,IC_(50))值分别为0.61、3.96、0.79 mg/mL。玉米谷蛋白水解物能够显著降低(P<0.05)乙醇诱导损伤的LO2细胞内活性氧(reactive oxygen species,ROS)的含量,并显著提高(P<0.05)超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽还原酶(glutathione reductase,GR)活性和还原型谷胱甘肽(glutathione,GSH)水平,对氧化应激损伤的LO2细胞具有保护作用。该研究为玉米谷蛋白水解物作为天然抗氧化剂应用在食品工业中奠定了理论基础。Corn glutenin hydrolysate(CGH)was prepared by hydrolyzing corn glutenin with protamex and alcalase.The antioxidant activity of CGH in vitro was studied.The oxidative damage model of LO2 cells induced by ethanol was used to study the antioxidant stress of CGH.The results showed that CGH had good antioxidant activity in vitro.When the protein concentration was 2 mg/mL,the scavenging rates of·OH and DPPH free radical were(86.69±0.022)%and(60.56±0.369)%,respectively,and the chelating capacity of Fe^(2+)was(91.58±0.025)%.CGH significantly decreased(P<0.05)the content of reactive oxygen species(ROS)in LO2 cells injured by ethanol,and significantly increased(P<0.05)the activities of superoxide dismutase(SOD)and glutathione reductase(GR),and the level of reduced glutathione(GSH)in LO2 cells injured by oxidative stress.This study laid a theoretical foundation for the application of CGH as a natural antioxidant in food industry.
