摘　　要：目的探讨circ_102476在胃癌中的表达及临床意义。方法从GEO数据库中下载circRNA数据集,包含3对胃癌组织和癌旁组织标本。采用R语言软件进行数据处理,获得差异表达circRNA。收集2014年3月-2019年3月苏州大学附属第一医院92例胃癌患者的胃癌组织及癌旁组织。应用qRT-PCR检测circ_102476,分析其在胃癌中的表达与临床因素的关系。用ROC曲线来验证circ_102476诊断胃癌的价值。结果筛选出99个差异表达基因,上调的circRNA有1个,下调98个,其中circ_102476下调最显著。qRT-PCR结果显示胃癌组织中circ_102476表达水平低于癌旁组织(P<0.05)。肿瘤晚期、分化差、发生远处转移是circ_102476表达下调相关因素。ROC曲线下面积为0.75。结论circ_102476在胃癌中表达下调,肿瘤分期、分化程度以及是否发生远处转移是表达下调的相关因素,可能在胃癌临床诊断中发挥作用。Objective To explore the expression of circ_102476 in gastric cancer and its clinical significance.Methods CircRNA datasets containing three pairs of adjacent tissue and gastric cancer tissue specimens were downloaded from GEO database.R language software was used for data processing to obtain differentially expressed circRNA.Gastric cancer tissues and adjacent tissues from 92 patients with gastrointestinal surgery gastric cancer in the First Affiliated Hospital of Soochow University from March 2014 to March 2019 were collected.QRT-PCR was used to detect circ_102476,and the relationship between its expression in gastric cancer and clinical factors was analyzed.ROC curve was used to verify the value of circ_102476 in the diagnosis of gastric cancer.Results 99 differentially expressed genes were screened out,with 1 up-regulated and 98 down-regulated circRNA.Among them,circ_102476 was down-regulated the most.QRT-PCR results showed the expression level of circ_102476 in gastric cancer tissues was lower than that in adjacent tissues(P<0.05).Advanced tumors,poorly differentiated,and distant metastasis were downregulated circ_102476 factors.The area under the ROC curve was 0.75.Conclusion Circ_102476 downregulated in gastric cancer.Tumor stage,degree of differentiation and whether distant metastasis related to the down-regulation of expression may play a role in the clinical diagnosis of gastric cancer.
