摘　　要：目的探讨维吾尔族乳腺癌中STAT3蛋白及mRNA的表达及其临床病理意义。方法收集维吾尔族乳腺癌组织80例,正常乳腺组织30例,运用RT-PCR和免疫组化法检测STAT3在不同分子分型维吾尔族乳腺癌中表达的差异及其与临床病理特征的关系;评估维吾尔族乳腺癌中STAT3蛋白及mRNA的表达情况及其与患者预后的关系。结果STAT3蛋白和mRNA在维吾尔族乳腺癌中的表达均高于正常组织(P<0.05);STAT3蛋白在不同分子分型维吾尔族乳腺癌组织中的表达差异具有统计学意义(P<0.05);STAT3蛋白表达与乳腺癌肿瘤大小、组织学分级、淋巴结转移、TNM分期均明显相关(P<0.05);在不同淋巴结转移情况、TNM分期乳腺癌组织中STAT3 mRNA的相对表达量比较差异均具有统计学意义(P<0.05)。绘制乳腺癌患者术后60个月生存曲线,STAT3低表达患者的中位生存期为54个月(95%CI:51.93~56.07),STAT3高表达患者的中位生存期为48个月(95%CI:42.21~53.79),STAT3低表达组患者的生存率明显高于高表达组(Log-rankχ^(2)=4.256,P=0.039)。结论STAT3高表达的乳腺癌可能更具有侵袭性,预后较差,其在维吾尔族乳腺癌发生发展过程中可能发挥着重要作用。Purpose To investigate the expression and clinicopathological significance of STAT3 protein and mRNA in breast cancer of Uygur nationality.Methods Eighty cases of breast cancer and thirty cases of normal breast tissue of Uygur nationality were collected.RT-PCR and immunohistochemistry were used to detect the STAT3 expression in different molecular types of breast cancer of Uygur nationality,and its relationship with clinicopathological features,and to evaluate the expression of STAT3 protein and mRNA in Uygur breast cancer and its relationship with the prognosis of patients.Results The expression of STAT3 protein and mRNA in Uygur breast cancer was significantly higher than that in normal tissue(P<0.05),the expression of STAT3 protein in different molecular types of Uygur breast cancer was different,with statistically significant(P<0.05),the expression of STAT3 protein was significantly correlated to tumor size,histological grade,lymph node metastasis and clinical stage(all P<0.05).There were significant differences among the relative expression of STAT3 mRNA in breast cancer tissues with respect to lymph node metastasis and TNM staging(all P<0.05).Survival curves of breast cancer patients after 60 months were drawn.The median survival time of patients with low STAT3 expression was 54 months(95%CI:51.93-56.07),and the median survival time of patients with high STAT3 expression was 48 months(95%CI:42.21-53.79).The overall survival rate of patients with low expression of STAT3 was significantly higher than that of patients with high expression of STAT3(Log-rankχ^(2)=4.256,P=0.039).Conclusion Breast cancer with high expression of STAT3 may be more invasive and have poor prognosis.Therefore,STAT3 may play an important role in the occurrence and development of breast cancer in Uygur nationality.
