摘 要:以2020年5月31日采自黑龙江省伊春市的柳叶芹(Czernaevia laevigata turcz.)为试验材料,利用十六烷基三甲基溴化铵法(CTAB)提取基因组DNA,采用Illumina NovaSeq平台进行高通量测序,得到柳叶芹完整的叶绿体基因组序列,分析柳叶芹叶绿体基因组组装、序列特征、系统发育。结果表明:柳叶芹叶绿体基因组总长度146161 bp,包括1个大单拷贝区长度为93538 bp、1个小单拷贝区长度为17779 bp,由2个反向重复序列分别分隔,每个反向重复序列为17422 bp。对柳叶芹叶绿体全基因组共鉴定出133个基因。系统发育分析表明,柳叶芹与疏叶当归有密切的亲缘关系。柳叶芹的叶绿体基因组序列,为研究伞形科其他植物提供了较为详细、完善的资料,为该物种鉴定以及群体和个体水平的遗传差异分析提供了新的方法。With Czernaevia laevigata turcz.collected from Yichun City,Heilongjiang Province on May 31,2020 as the test material,the genomic DNA was extracted by the cetyltrimethylammonium bromide method(CTAB).Through throughput sequencing,the complete chloroplast genome sequence was obtained,and the chloroplast genome assembly,sequence characteristics and phylogeny were analyzed.The total length of the chloroplast genome was 146161 bp,including a large single-copy region of 93538 bp and a small single-copy region of 17779 bp,separated by two inverted repeats,each of which was 17422 bp.A total of 133 genes were identified from the whole chloroplast genome of willow celery.By phylogenetic analysis,celery and Angelica sparse have close kinship.The chloroplast genome sequence of willow celery would provide more detailed and complete information for the study of other plants of the Umbelliferae,and a new method for the identification of this species and the analysis of genetic differences at the population and individual levels.
