摘 要:目的研究肾透明细胞癌与癌旁组织的基因表达差异,探讨差异表达基因在肾透明细胞癌发生发展中的作用。方法高通量测序分析肾透明细胞癌组织和癌旁组织的基因差异表达;实时荧光定量PCR及Western blot验证与代谢相关的基因。结果生物信息学显示肾透明细胞癌中有40个ATP合成酶相关基因表达增加,实时荧光定量PCR及Western blot验证了部分二代测序结果,与癌旁组织比较,肾透明细胞癌的ATP5F1 mRNA的相对丰度及蛋白表达增加(P<0.05)。结论肾透明细胞癌可以导致多种基因表达变化,其中ATP合成酶表达增加可能在肾透明细胞癌的发生发展中起到重要作用。Objective To explore the difference of gene expression between clear cell renal cell carcinoma and adjacent tissues,and to explore the role of differentially expressed genes in the occurrence and development of clear cell renal carcinoma.Methods High-throughput sequencing was used to analyze clear cell renal cell carcinoma and adjacent tissues.The genes related to metabolism were verified by real-time fluorescence quantitative PCR and Western blot.Results Bioinformatics showed that the expression of 40 ATP synthase-related genes in clear cell renal carcinoma increased.Real-time PCR and Western blot verified this result.Compared with adjacent tissues,the relative abundance of ATP5F1 mRNA and protein expression of ccRCC increased(P<0.05).Conclusion Clear cell renal carcinoma may lead to changes in the expression of a variety of genes,among which the increased expression of ATP synthase may play an important role in the occurrence and development of clear cell renal cell carcinoma.
