摘 要:用MTT法检测羌活有效活性成分阿魏酸对PK-15细胞的安全浓度,采用3种给药方法优化最佳给药方式,梯度给药优化最佳抗病毒增殖浓度及作用时间,用RT-qPCR检测炎症因子和细胞因子的表达水平。结果显示,羌活有效活性成分阿魏酸对PK-15细胞的最高安全浓度为160μmol/mL。最佳给药时间是猪细小病毒(PPV)感染细胞4 h后,且羌活有效活性成分阿魏酸呈剂量依赖性关系显著降低PPV的病毒表达;与空白组相比,羌活有效活性成分阿魏酸显著降低PPV感染PK-15诱发IL-1β、IL-2、IL-8、IL-6、IL-12、IL-18和TNF-α的表达,显著升高PPV感染PK-15诱发IL-17的表达。表明羌活有效活性成分阿魏酸抑制PPV在PK-15细胞中的增殖,降低PPV感染细胞激活炎症因子的表达水平,有良好的抗炎和抗病毒活性。MTT assay was used to detect the safe concentration of ferulic acid,an active component of Notopterygium incisum,on PK-15 cells.Three methods were used to optimize the optimal administration method.Under the optimal administration mode,gradient administration was used to optimize the optimal antiviral proliferation concentration and action time,and the expression levels of inflammatory factors and cytokines were detected by RT-qPCR.The results showed that the highest safe concentration of ferulic acid was 160μmol/mL.Compared with the blank group,ferulic acid could significantly decrease the expressions of IL-1β,IL-2,IL-8,IL-6,IL-12,IL-18 and TNF-αinduced by PK-15,and increase the expression of IL-17 induced by PK-15.The results showed that ferulic acid inhibited the proliferation of PPV in PK-15 cells,and reduced the expressions of inflammatory factors in PPV infected cells.
