褪黑素对鱼藤酮致帕金森病大鼠运动功能和炎症反应的影响

摘  要:目的:研究褪黑素(melatonin,MT)对鱼藤酮所导致的帕金森病(parkinson’s disease,PD)大鼠的运动功能和炎症反应的影响。方法:选取2020年6月—2021年7月齐齐哈尔医学院动物实验中心实验动物中健康清洁的SPF级雄性Wistar大鼠,共36只。建立PD模型大鼠:将大鼠随机分为三组,分别为:对照组、PD造模组、PD+褪黑素组。分组后,对PD造模组以及PD+褪黑素组施加鱼藤酮皮下给药36d,对照组给予等量0.9%氯化钠溶液。之后按标准对各组大鼠进行行为学评分。造模成功后,PD+褪黑素组大鼠注射褪黑素14d,PD造模组和对照组给予等量0.9%氯化钠溶液。在上述干预后,采用旷场实验以及转棒实验对各组大鼠进行行为学实验。最后取脑,使用ELISA法检测大鼠黑质N F-κB、TH、纹状体IL-1B、IL-6、TNF-α的活性。结果:与对照组比较,PD造模组大鼠均出现典型的PD样症状,运动功能各项指标有所下降;经检测TH蛋白在黑质中的表达下降,NF-κB在黑质中的表达增加,TNF-α和IL-1β、IL-6在纹状体中的表达增加;PD+褪黑素组大鼠经干预后,相较造模组P D样症状和运动功能各项指标均得到改善,经检测T H蛋白在黑质中的表达增加,NF-κB在黑质中表达减少,TNF-α、IL-1β、IL-6在纹状体中的表达减少。结论:褪黑素可以通过改善PD大鼠运动功能和炎症反应产生一定的保护作用。Objective To study the effect of Melatonin(MT)on the motor function and inflammatory response of Rotenoneinduced rat model of Parkinson’s disease.Methods Thirty-six SPF grade male Wistar rats were selected from The Animal Experiment Center of Qiqihar Medical College from June 2020 to July 2021.Establishment of PD model rats:Rats were randomly divided into three groups:control group,PD modeling group and PD+melatonin group.After grouping,the PD modeling group and PD+melatonin group were subcutaneously administered rotenone for 36 days,and the control group was given the same amount of normal saline.Then the rats in each group were given behavioral score according to the standard.After modeling,rats in PD+melatonin group were injected with melatonin for 14 days,and rats in PD modeling group and control group were given the same amount of normal saline.After the intervention,open field experiment and rotary rod experiment were used to conduct behavioral experiments.Finally,the activities of NF-κB,TH,IL-1B,IL-6 and TNF-αin substantia nigra and striatum were detected by ELISA.Results Compared with the control group,PD model rats all had typical PD-like symptoms,and various indexes of motor function decreased;The expression of TH protein in substantia nigra decreased,and NF-κB in substantia nigra increased.The expression of TNF-α,IL-1βand IL-6 in the striatum increased.Compared with the PD module group,the PD-like symptoms and various indexes of motor function were improved.Expression of TH protein in substantia nigra increased,the expression of NF-κB in substantia nigra decreased,and the expression of TNF-α,IL-1β,and IL-6 in the striatum decreased.Conclusion Melatonin can have a certain protective effect by improving the motor function and inflammatory response in Rotenone-induced PD rats.
