摘　　要：目的介绍一种稳定可靠的肝组织上和体外培养的大鼠肝窦内皮细胞(liver sinusoidal endothelial cells,LSECs)的扫描电子显微镜样本制作方法,以便于形态学方面的研究。方法采用戊二醛、单宁酸、锇酸三步固定;酒精梯度脱水;六甲基二硅氮烷置换;离子喷镀导电处理。结果扫描电子显微镜下可观察到肝组织上及体外培养的大鼠LSECs的精细结构。结论该实验方法可作为可靠的大鼠LSECs扫描电子显微镜样本的制作方法。Objective To introduce a stable and reliable scanning electron microscope sample preparation method of rat liver sinusoidal endothelial cells(LSECs)cultured in liver tissue and in vitro to facilitate the study of morphology.Methods Three-step fixation of glutaraldehyde,tannic acid and osmium acid was used;alcohol gradient dehydration;hexamethyldisilazane replacement;ion spraying conductive treatment.Results The fine structure of rat LSECs cultured in vitro could be observed under the scanning electron microscope.Conclusion This experimental method can be used as a reliable method for preparing rat liver sinusoidal endothelial cells scanning electron microscope samples.
