摘  要:目的运用网络药理学方法探讨盐酸小檗碱治疗抑郁症的作用机制。方法通过中药系统药理学数据库与分析平台及PharmMapper网站预测盐酸小檗碱的作用靶点,并利用STRING数据库构建靶点的蛋白-蛋白相互作用(PPI)网络,获得盐酸小檗碱的潜在作用靶点。检索GeneCards数据库以获得抑郁症相关靶点,与盐酸小檗碱的潜在作用靶点取交集后,得到盐酸小檗碱治疗抑郁症的潜在作用靶点,然后构建PPI网络并进行拓扑结构分析,获得核心潜在作用靶点。通过DAVID数据库对盐酸小檗碱治疗抑郁的潜在作用靶点进行基因本体论(GO)功能富集分析及京都基因和基因组百科全书(KEGG)通路富集分析。结果共获得267个盐酸小檗碱的潜在作用靶点和14271个抑郁症相关靶点,取交集后,共获得盐酸小檗碱治疗抑郁症的潜在作用靶点184个,其中ALB、HSP90αA1、CYCS、CAT、ESR1、CDK2和G6PD为盐酸小檗碱治疗抑郁的核心潜在作用靶点。GO生物学功能富集分析结果显示,盐酸小檗碱治疗抑郁症的184个潜在作用靶点参与细胞增殖分化、细胞凋亡、对细胞周期的调节、缺氧的调控等24个生物学过程;KEGG通路富集分析结果显示,盐酸小檗碱治疗抑郁症的184个潜在作用靶点能够对p53信号通路、叉头框蛋白O(FoxO)信号通路等18个KEGG通路进行调节。结论盐酸小檗碱可能通过p53信号通路、FoxO信号通路、调控缺氧等多种途径发挥治疗抑郁症的作用。Objective To investigate the mechanism of action of berberine hydrochloride in the treatment of depression using the network pharmacology method.Methods The action targets of berberine hydrochloride were predicted through the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform and the PharmMapper website,and STRING database was employed to construct a protein-protein interaction(PPI)network of the targets to obtain the potential action targets of berberine hydrochloride.The depression-related targets were obtained through searching the GeneCards database and were intersected with the potential action targets of berberine hydrochloride.After the intersection,the potential action targets of berberine hydrochloride in the treatment of depression had been gained,a PPI network was constructed,a topological structure analysis was performed,and the core potential action targets were obtained.Gene Ontology(GO)functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were performed on the potential action targets of berberine hydrochloride in the treatment of depression via DAVID database.Results A total of 267 potential action targets of berberine hydrochloride and 14271 depression-related targets were obtained.After the intersection,184 potential action targets of berberine hydrochloride in the treatment of depression were obtained,including ALB,HSP90αA1,CYCS,CAT,ESR1,CDK2,and G6PD which were the core potential action targets of berberine hydrochloride in the treatment of depression.The results of GO functional enrichment analysis on the biological process showed that 184 potential action targets of berberine hydrochloride in the treatment of depression were involved in 24 biological processes,including cell proliferation and differentiation,apoptosis,cell cycle regulation,and hypoxia regulation and control.The results of KEGG pathway enrichment analysis revealed that 184 potential action targets of berberine hydrochloride in the treatment o
