佛手和代代酸橙授粉对香橼果形的花粉直感效应研究

摘  要:佛手(Citrus medica L.var.sarcodactylis Swingle)、香橼(Citrus medica L.)与代代酸橙(C.aurantium var.daidai Tanaka)皆为芸香科柑橘属植物,三者在果形上存在极大差异.预实验发现3个物种近距离栽种,大部分香橼果形发生变异.为探明柑橘属花粉直感改变果形的机制,以上述3种花粉为父本,以香橼为母本进行授粉,从花芽到果实阶段进行形态学数据统计与观察,并收集3种花粉及开花前7天、开花授粉当天及授粉后7天3个时期的香橼雌蕊样本进行荧光定量PCR.结果表明:从形态上观察到佛手、代代酸橙的授粉能使香橼的果形指数发生变化,果皮和果形均与香橼不同;对CNR,CRC,SUN,KAN2,YABBY2,YABBY5等6个基因mRNA的表达进行定量分析,发现它们的表达模式与香橼果形变化密切相关.根据上述结果,佛手和代代酸橙的花粉授粉能够有效改变香橼的果形,果形的改变可能与上述果形基因的表达变化有关.The fingered citron(Citrus medica L.var.sarcodactylis Swingle),the citron(Citrus medica L.)and Daidai(C.aurantium var.Daidai Tanaka)were all classified as citrus plants of the Rutaceae family,and there were great differences among them in fruit shape.In the pre-experiment,it was found that most of the citron fruit shape varied when the 3 species were planted closely.In order to investigate the pollen xenia effect on fruit shape development of citron,the pollen of fingered citron,citron and Daidai were used as male parent and citron as the female parent for pollination.The morphological characters of the pollinated flower were monitored until the mature fruit stage.All the pollen and the citron pistils from 7 days before flowering,the day of flowering and 7 days after flowering were collected to perform qRT-PCR.Morphologically,it was observed that the pollination of citron and Daidai changed the fruit shape index of citron,which showed the peel and the fruit shape were different from the citron.The mRNA expression analysis of CNR,CRC,SUN,KAN2,YABBY2 and YABBY5 genes revealed that these genes might be closely related to the fruit shape variation of citron.According to the results,it was concluded that pollination of fingered citron and Daidai pollen could significantly change the fruit shape of citron,which might be related to the fruit-shaped genes expression.
