摘 要:为了解猪链球菌2型菌株对昆明小鼠的致病性,并建立其感染昆明小鼠模型,本试验分别取2头疑似患链球菌病的病死猪脑组织、血液分别涂布于含5%胎牛血清的TSA固体培养基中进行细菌分离;并对分离获得的细菌进行形态观察、PCR鉴定、药物敏感性试验、小鼠致病性试验及组织病理切片观察。结果显示,分离菌为革兰氏阳性短链球菌,经PCR鉴定为猪链球菌2型,命名为LY株;药敏试验证实LY株对青霉素、红霉素、氯霉素、苯唑西林、环丙沙星、诺氟沙星敏感,对四环素、多粘菌素B、克林霉素不敏感;LY株对小鼠致死剂量为2.2×10^(9) cfu/只;组织病理切片观察发现小鼠肺脏、脾脏及心肌均出现不同程度的病变。本研究明确了山东省猪链球2型毒株的毒力,为疫苗候选菌株的筛选提供了理论依据。In order to understand the pathogenicity of Streptococcus suis type 2 strain to Kunming mice and establish its infection model,the brain tissue and blood of two dead pigs suspected of S.suis were coated in TSA solid medium containing 5%fetal bovine serum for bacterial isolation;the isolated bacterum was identified by morphology observation,PCR identification,drug sensitivity test,mouse pathogenicity test and histopathological section observation.The results showed that the isolated bacterum belonged to Gram-positive Streptococcus,which was identified as S.suis type 2 by PCR and named LY strain.The drug sensitivity test showed the LY strain was sensitive to penicillin,erythromycin,chloramphenicol,oxacillin,ciprofloxacin and norfloxacin,but resistant to tetracycline,polymyxin B and clindamycin;the lethal dose to one mice was 2.2×10^(9) cfu.The tissue section observation found that there were different degrees of lesions in lung,spleen and myocardium.This study clarified the virulence of S.suis type 2 in Shandong Province,and provided a theoretical basis for screening of vaccine candidate strains.
