摘  要:目的观察miRNA-26b(miR-26b)在野生型C57/BL6J小鼠和瘦素基因遗传性缺陷ob/ob小鼠组织中的表达特征,以及瘦素对3T3-L1成熟脂肪细胞中miR-26b表达的调控。方法取16周龄C57/BL6J小鼠和ob/ob小鼠的心脏、肝脏、脾、胰、肾脏、肺、小肠、骨骼肌、白色脂肪组织和棕色脂肪组织,采用实时荧光定量聚合酶链反应(qRT-PCR)技术检测小鼠各种组织中miR-26b表达水平;采用100 ng/mL的瘦素干预3T3-L1成熟脂肪细胞16 h,采用qRT-PCR技术检测细胞中miR-26b表达水平。结果ob/ob小鼠的体质量、血糖、甘油三酯及总胆固醇水平明显高于C57/BL6J小鼠,差异均有统计学意义(P<0.05)。miR-26b在C57/BL6J小鼠心脏、脂肪、胰组织中表达水平较高,而在小肠、肾脏、骨骼肌组织中表达水平较低。与C57/BL6J小鼠比较,miR-26b在ob/ob小鼠心脏、胰、白色脂肪组织中表达水平明显下降,而在肺、骨骼肌、肾脏及小肠组织中表达水平升高,差异有统计学意义(P<0.05);在小鼠3T3-L1成熟脂肪细胞中,瘦素可明显下调miR-26b表达。结论ob/ob小鼠各种组织中miR-26b表达呈现不同的变化特征,miR-26b是一种与机体糖脂代谢功能密切相关的miRNA。Objective To observe the expression of miRNA-26b(miR-26b)in various tissue in C57/BL6J mice and ob/ob mice and the regulative roles of leptin on miR-26b in 3T3-L1 mature adipocytes.Methods The level of miR-26b in heart,liver,spleen,pancreas,kidney,lung,small intestine,skeletal muscle,white adipose and brown adipose tissue were determined in 16-week-old C57/BL6J mice and ob/ob mice using real-time quantitative reverse transcription polymerase chain reaction(qRT-PCR)technique.The 3T3-L1 mature adipocytes were treated with 100 ng/mL leptin for 16 hours.The level of miR-26b in adipocytes was detected by qRT-PCR.Results Body mass,blood glucose,triglyceride,total cholesterol in ob/ob mice were significant higher than those in C57/BL6 mice(P<0.05).The heart,adipose,pancreas tissue of C57/BL6J mice displayed high miR-26b level,while the small intestine,kidney,skeletal muscle tissue had low miR-26b level.The expression levels of miR-26b decreased significantly in heart,pancreas,white adipose tissue while increased in lung,skeletal muscle,kidney,small intestine of ob/ob mice compared to C57/BL6J mice(P<0.05).Leptin could dramatically down-regulated miR-26b expression in 3T3-L1 mature adipocytes.Conclusion The miR-26b expression in ob/ob mice exhibited features of the various tissue.miR-26b might be involved in the regulation of glucose and lipid metabolism in mice.
