摘  要:目的考察桑寄生不同提取部位对黄嘌呤氧化酶的体外抑制活性,以及对高尿酸症小鼠的降尿酸作用。方法通过萃取获得桑寄生石油醚(PE),乙酸乙酯(EA),正丁醇(NB)和水(W)部位。利用UV检测各部位对黄嘌呤氧化酶的抑制活性,利用SPSS 20.0软件来计算半数抑制浓度(IC;),判断其抑制类型。取KM小鼠随机分成正常组,模型组,别嘌醇组(10 mg·kg^(-1))和4个给药组(PE组,EA组,NB组,W组,6.9 g·kg^(-1),按生药量计),每组8只。除正常组外,各组每日给予氧嗪酸钾(250 mg·kg^(-1),i.g.),以复制高尿酸小鼠模型;1 h后,正常组与模型组给予等体积0.5%CMC-Na(i.g.),其余各组给予相应药物(i.g.),连续7 d。末次给药1 h后采用ELISA检测血清中黄嘌呤氧化酶(XOD),尿酸(UA),肌酐(Cr)和尿素氮(BUN)水平。结果桑寄生PE,EA,NB和W部位的IC_(50)分别为338,73,67和1424μg·mL^(-1),EA和NB部位的体外抑制XOD活力最强,其抑制类型为可逆性抑制。动物实验结果显示,模型组小鼠血清UA,XOD,Cr和BUN水平显著提高(P<0.05,P<0.01);与模型组小鼠比较,别嘌醇组和EA组均能显著降低上述指标(P<0.05,P<0.01)。结论桑寄生EA和NB部位具有较强的体外抑制XOD活性,其EA部位经体内代谢能有效降低小鼠高尿酸水平,其治疗作用可能与改善肾功能相关。Objective To investigate the inhibitory activity of different Taxillus chinensis extracts on xanthine oxidase in vitro,and the uric acid-lowering effect on hyperuricemia mice.Methods Taxillus chinensis petroleum ether(PE),ethyl acetate(EA),n-butanol(NB)and water(W)fractions were obtained by extraction.The inhibitory activity of each Taxillus chinensis extracts to xanthine oxidase was detected by UV,and half inhibitory concentration(IC;)was calculated by SPSS 20.0 and determine the type of inhibition.KM mice were randomly divided into normal group,model group,allopurinol group(10 mg·kg^(-1))and 4 dose groups(PE group,EA group,NB group,W group,6.9 g·kg^(-1),by raw material),with 8 mice in each group.Except for normal group,other groups were given potassium oxonate(250 mg·kg^(-1),i.g)to induce hyperuricemia model,1 h after modeling,the normal group and model group were given 0.5% CMC-Na(i.g)at the same volume,and the other groups were given corresponding drugs(i.g)for 7 consecutive days.One hour after last administration,serum levels of xanthine oxidase(XOD),uric acid(UA),creatinine(CR)and urea nitrogen(BUN)were determined by ELISA.Results The IC_(50) of PE,EA,NB and W parts of Taxillus chinensis were 338,73,67 and 1424μg·mL^(-1).The inhibitory activity of XOD was strongest in EA and NB parts,and the inhibitory type was reversible inhibition.The results of animal experiments showed that the levels of serum UA,XOD,Cr and BUN in model group were significantly increased(P<0.05,P<0.01);Compared with model mice,the above indexes were significantly decreased in allopurinol and EA group(P<0.05,P<0.01).Conclusion The EA and NB parts of Taxillus chinensis have strong XOD inhibition activity in vitro,but only EA parts of Taxillus chinensis can effectively reduce the level of hyperuric acid in mice after metabolism in vivo,and its therapeutic effect may be related to the improvement of renal function.
