摘  要:目的探讨癫痫大鼠颞叶、海马区多药耐药基因1(MDR1)及P-糖蛋白(Pgp)的表达及其相互关系。方法选择健康清洁级雄性SD大鼠50只。采用随机数字表法将大鼠分为观察组和对照组。观察组大鼠海马区注射海人酸(KA)2μl(含KA 1μg),制作颞叶耐药性癫痫(RE)模型(成功22只),对照组同法注射0.9%氯化钠注射液2μl。两组大鼠饲养、观察1个月后,分别取颞叶、海马区组织,采用免疫组化及实时荧光定量PCR法检测MDR1 mRNA和Pgp表达水平,并分析其相互关系。结果观察组颞叶、海马区的MDR1 mRNA、Pgp表达水平均高于对照组,差异均有统计学意义(均P<0.05)。观察组颞叶、海马区MDR1 mRNA表达水平与Pgp表达水平呈正相关(r=0.55,P<0.05)。结论MDR1、Pgp可能参与RE的发病机制,可为开发相关新的治疗方法或药物提供依据。Objective To investigate the expression and relationship between MDR1 gene and Pgp in temporal lobe and hippocampus of rats with kainic acid-induced epilepsy.Methods A total of fifty SD rats were divided into observation group and control group by random number table method.The rats in the observation group were injected with 2μl KA(including 1μl KA1)in the hippocampus,and the model of drug-resistant epilepsy(RE)in the temporal leaf was established(22 rats were successfully).The rats in the control group were injected with 0.9% sodium chloride injection 2μl.The expression of MDR1 mRNA and Pgp in temporal lobe and hippocampus was detected with real-time quantitative RT-PCR and immunohistochemistry,respectively,after observing for 1 month.Results The expression levels of MDR1 mRNA and Pgp in the temporal lobe and hippocampus in epileptic group were significantly higher than those in control group(all P<0.05),and the expression level of MDR1 mRNA was positively correlated with Pgp(r=0.55,P<0.05).Conclusion MDR1 and Pgp are up-regulated in the temporal lobe and hippocampus of rats with kainic acid-induced epilepsy,suggesting they might be involved in the pathogenesis of drug-resistant epilepsy.
