摘　　要：目的探讨细胞周期激酶抑制因子4基因座中反义非编码RNA(ANRIL)在急性髓细胞性白血病(AML)中的表达及与患者临床特征的关系。方法选取82例AML患者,作为观察组,依据患者病情分为初治组(n=53)、缓解组(n=19)和复发组(n=10),另选取31例非血液病健康者,作为对照组。采用实时荧光聚合酶链反应(PCR)检测各组受试者骨髓ANRIL m RNA的相对表达量,并分析其与AML患者临床特征的关系。结果观察组、初治组、缓解组、复发组患者骨髓ANRIL mRNA的相对表达量均明显高于对照组(P﹤0.01)。复发组患者骨髓ANRIL m RNA相对表达量明显高于缓解组和初治组(P﹤0.01),缓解组患者骨髓ANRIL mRNA的相对表达量明显低于初治组(P﹤0.01)。血小板计数﹤20×10^(9)/L、疾病进展、预后未缓解或缓解后再复发AML患者骨髓ANRIL mRNA相对表达量分别明显高于血小板计数≥20×10^(9)/L、疾病无进展、预后缓解患者(P﹤0.01)。结论ANRIL在AML患者中高表达,其表达水平能反映患者的疾病严重程度,可指导临床诊疗。Objective To investigate the expression of antisense non-coding RNA in the inhibitors of cyclin-dependent kinase 4 locus(ANRIL)in acute myelogenous leukemia(AML)and its relationship with the clinical characteristics of patients.Method A total of 82 AML patients were selected as the observation group,and were divided into the initial treatment group(n=53),the response group(n=19)and the relapse group(n=10)according to their condition.Another 31healthy people without blood disease were selected as the control group.The relative expression of ANRIL mRNA in the bone marrow of subjects in each group was detected by the real-time fluorescence polymerase chain reaction(PCR),and its relationship with the clinical features of AML patients was also analyzed.Result The relative expression of ANRIL mRNA in the bone marrow of the observation group,initial treatment group,response group,and relapse group were significantly higher than that of the control group(P<0.01).The relative expression of ANRIL mRNA in the bone marrow of the patients in the relapse group was significantly higher than that in the response group and the initial treatment group(P<0.01).The relative expression of ANRIL m RNA in the bone marrow of patients with platelet count<20×10^(9)/L,disease progression,no response in prognosis or relapse after response AML patients were significantly higher than that of patients with platelet count≥20×10^(9)/L,disease progression-free,and prognosis response(P<0.01).Conclusion ANRIL is up-regulation in AML patients,and its expression level could reflect the disease severity of patients and could be used for clinical diagnosis and treatment guidance.
