摘  要:目的观察齿龈内阿米巴(E.g)接种对大鼠牙周炎发生发展的影响。方法将24只雄性SD大鼠随机分为对照组(C组)、齿龈内阿米巴干预组(E.g组)、实验性牙周炎组(EP组)、混合干预组(mix组),每组6只。C组给予醋酸泼尼松龙肌注,同时生理盐水涂抹于大鼠左上第一磨牙龈缘。E.g组给予醋酸泼尼松龙肌注,同时E.g悬液涂抹于大鼠左上第一磨牙龈缘进行E.g接种。EP组采用4-0丝线结扎左侧上颌第一磨牙制作牙周炎模型。mix组在制作牙周炎模型的基础上给予醋酸泼尼松龙肌注,同时E.g悬液涂抹左上第一磨牙龈缘。从实验第1天开始至第14天,每日称量大鼠体质量,然后用牙周探针测量左上颌第一磨牙的牙周袋探诊深度(PD),观察并记录龈沟出血指数(SBI)。实验第15天,处死大鼠后获取左侧上颌骨标本,使用microCT观察组大鼠左上颌第一磨牙牙槽骨吸收情况,HE染色后观察大鼠左上颌第一磨牙牙周组织病理学变化。取大鼠腹主动脉血,采用ELISA法检测大鼠血清中环氧合酶2(COX2)、基质金属蛋白酶9(MMP9)、白细胞介素6(IL-6)、血管内皮生长因子(VEGF)。结果C组和E.g组大鼠体质量稳步增长,而EP组与mix组大鼠体质量均在实验后期有较明显的增长。EP组与mix组PD在第10~12天左右达到高峰并趋于稳定,E.g组在实验第5~7天牙龈肿胀明显,龈缘平面高于正常龈缘。E.g组、EP组与mix组大鼠左上颌第一磨牙的SBI在前7 d均有不断增加,但EP组与mix组牙龈出血情况明显重于E.g组。C组大鼠左上颌第一磨牙牙槽骨无吸收,EP组、E.g组和mix组牙周膜增宽明显,并且EP组、mix组均可见牙槽骨吸收,mix组较EP组牙槽骨吸收更严重。C组大鼠牙龈无炎症浸润,E.g组大鼠可见龈沟内有明显炎症浸润,EP组与mix组大鼠牙周组织可见大面积炎症浸润。与C组相比,E.g组大鼠血清中MMP9、IL-6、VEGF表达水平显著升高(P均<0.05),EP组、mix组大鼠血清中COX2�Objective To observe the effect of Entamoeba gingivalis(E.g) inoculation on the development and progression of periodontitis in rats.Methods Twenty-two male SD rats were randomly divided into the control group(group C),gingival Entamoeba gingivalis intervention group(E.g group),experimental periodontitis group(EP group),and mixed intervention group(mix group),with 6 rats in each group.Rats in the group C were given intramuscular injection of prednisolone acetate,and normal saline was smeared on the gingival margin of the left upper first molar of rats.The rats in the E.g group were given intramuscular injection of prednisolone acetate,and E.g suspension was smeared on the gingival margin of the left upper first molar for E.g inoculation.In the EP group,the periodontitis model was made by ligating the left maxillary first molar with 4-0 silk thread.In the mix group,prednisolone acetate was injected intramuscularly on the basis of periodontitis model,and E.g suspension was smeared on the gingival margin of the left upper first molar.From the first day to the 14 th day of the experiment,the body weight of the rats was weighed every day,and then the periodontal probe was used to measure the periodontal pocket probing depth(PD) of the left maxillary first molar,and the gingival sulcus bleeding index(SBI) was observed and recorded.On the 15 th day of the experiment,the left maxillary specimens were obtained after the rats were killed.The alveolar bone resorption of the left maxillary first molars of the rats was observed by the microCT.The periodontal histopathological changes of the left maxillary first molars were observed after HE staining.The serum levels of cyclooxygenase-2(COX2),matrix metalloproteinase-9(MMP-9),interleukin-6(IL-6)and vascular endothelial growth factor(VEGF) were detected by ELISA.Results The body weight of rats in the groups C and E.g increased steadily,while the body weight of rats in the EP group and mix group increased significantly in the later stage of the experiment.The PD of the EP group
