摘 要:目的运用网络药理学分析方法,初步探讨三七配伍生蒲黄抗糖尿病视网膜病变(DR)的潜在作用机制。方法通过TCMSP检索并筛选三七与生蒲黄的活性成分及药物靶点。借助多个数据库检索获得DR相关靶点。通过Venny2.1.0工具获得药物与疾病的交集靶点。利用Cytoscape3.7.2软件绘制蛋白互作网络图及药物-活性成分-靶点-疾病网络图,并进行网络拓扑学分析;利用R软件Bioconductor的工具包进行基因本体学(GO)功能分析和Kyoto基因与基因组百科全书(KEGG)通路分析,得到关键靶点及潜在作用通路。结果筛选出三七与生蒲黄中148个化合物、330个相应作用靶点,预测出与DR相关靶点159个,GO功能富集分析得到93个关键基因,KEGG通路富集分析得到154条信号通路。结论三七配伍生蒲黄防治DR的作用机制,主要涉及GAPDH、INS、CAT、IL-6、VEGFA等靶点基因,可能通过调控AGE-RAGE、HIF-1等信号通路,改善DR的糖/脂/蛋白代谢异常、氧化应激以及炎症反应。OBJECTIVE To preliminarily explore the underlying mechanism of antidiabetic retinopathy(DR)in Panax Notoginseng combined with Pollen Typhae with network pharmacology analysis.METHODS The active ingredients and drug targets of Panax Notoginseng and Pollen Typhae were retrieved and screened by TCMSP.DR-related targets were obtained by means of multiple database searches.The intersecting targets of drugs and diseases were obtained by Venny2.1.0 tool.Protein-protein interaction network diagram and drug-active ingredient-targetdisease network diagram was drawn by Cytoscape 3.7.2 software,and network topology analysis was carried out;Gene ontology(GO)function analysis and Kyoto encyclopedia gene and genome(KEGG)pathway analysis were carried out by R software Bioconductor toolkit,and the key targets and potential action pathways were obtained.RESULTS A total of 148 compounds and 330 corresponding targets were screened out from Panax Notoginseng and Pollen Typhae,159 targets related to DR were predicted,93 key genes were obtained from GO functional enrichment analysis,and 154 signa ling pathways were obtained from KEGG pathway enrichment analysis.CONCLUSIONS Mechanisms of action of Panax notoginseng combined with Pollen Typhae protects against DR mainly involves GAPDH,INS,CAT,IL-6,VEGFA and other target genes.By regulating AGE-RAGE,HIF-1 and other signal pathways,it improves the abnormal glucose,lipid,protein metabolism,oxidative stress and inflammatory reaction of DR.
