摘  要:以持续供给正十六烷为外界环境压力,从大庆石油污染土壤中筛选出一株正十六烷高效降解菌株,命名为L1.经菌株形态特征观察和16S rDNA基因测序,确定该降解菌株为溶血不动杆菌(Acinetobacter haemolyticus).通过单因素试验探究环境因子(正十六烷体积分数、菌液接种量、初始pH值、NaCl质量浓度、培养时间)对该菌株降解正十六烷能力的影响.研究结果表明,该菌株在接种量4%、初始pH值8、NaCl质量浓度5 g/L,35℃,150 r/min振荡培养的条件下,12 d后对体积分数为0.5%(3867 mg/L)的正十六烷降解率可达62.0%.In this paper, the continuous supply of n-hexadecane was taken as the external environmental pressure, and a strain of n-hexadecane degrading efficiently was selected from Daqing petroleum-contaminated soil and named L1. Observation of the morphological characteristics of the strain and 16 S rDNA gene sequencing confirmed that the degrading strain was Acinetobacter haemolyticus. A single factor experiment was conducted to explore the effects of environmental factors(n-hexadecane volume fraction, inoculum amount, initial pH, NaCl mass concentration, culture time) on the ability of the strain to degrade n-hexadecane. The results of the study showed that the strain had an inoculum volume of 4%, initial pH value of 8, NaCl mass concentration of 5 g/L, 35 ℃, and 150 r/min shaking culture conditions, The degradation rate of n-hexadecane with volume fraction of 0.5%(3 867 mg/L) reached 62.0% after 12 days.
