摘 要:心搏骤停、缺血性脑卒中、休克和严重烧伤等均可出现脑缺血-再灌注(ischemia/reperfusion,I/R)损伤,其病理生理机制错综复杂,预后差。铁死亡作为一种由铁依赖性的脂质过氧化物和活性氧(ROS)过度累积引起的新型程序性细胞死亡模式,主要涉及谷胱甘肽的耗竭、铁代谢异常和脂质过氧化。已有研究表明,铁死亡参与脑I/R损伤的发生发展,且渐渐成为研究的热点。本文从铁死亡概述、铁死亡与脑I/R损伤和铁死亡调控脑I/R损伤的机制进展三方面展开,为进一步探讨脑I/R损伤相关疾病的病理生理机制和寻找新的治疗靶点提供参考。Cerebral ischemia/reperfusion(I/R)injury can occur in cardiac arrest,ischemic stroke,shock and severe burns.The pathophysiological mechanism is complicated and the prognosis is poor.Ferroptosis,as a new type of programmed cell death caused by iron-dependent lipid peroxides and excessive accumulation of reactive oxygen species(ROS),mainly contains glutathione depletion,iron metabolism abnormal,and lipid peroxidation.It has been known that ferroptosis is involved in the development of cerebral I/R injury,and has gradually become a research hotspot.This article reviews the overview of ferroptosis,ferroptosis and cerebral I/R injury,and the molecular mechanisms of ferroptosis during cerebral I/R injury,so as to provide some references for further exploring the pathophysiological mechanisms of several fatal disease related to cerebral I/R injury and finding new therapeutic targets.
