兰尼碱受体2在浸润性乳腺癌组织中的表达及其临床病理学意义

摘　　要：目的:探讨兰尼碱受体2(ryanodine receptor 2,RyR2)在浸润性乳腺癌组织中的表达及其临床病理学意义。方法:使用聚合酶链式反应、蛋白质印迹、免疫组织化学染色等方法检测RyR2在浸润性乳腺癌患者乳腺癌、乳腺癌旁和正常乳腺组织样本中的表达水平,并分析其与乳腺癌组织学分级、淋巴结转移和超声检查特征间的关系。结果和结论:RyR2在浸润性乳腺癌组织中的表达水平显著低于乳腺癌旁和正常乳腺组织(均P<0.01),且其在浸润性乳腺癌组织中的低表达与乳腺癌的组织学分级高、雌激素受体阴性、存在微钙化和淋巴结转移相关(均P<0.05)。结合超声检查和RyR2检测结果能为浸润性乳腺癌的诊断提供更准确的判读结果。Objective:To investigate the expression of ryanodine receptor 2(RyR2)in invasive breast cancer tissues and its clinicopathological significance.Methods:RyR2 expression levels in breast,adjacent and normal breast tissue samples from patients with invasive breast cancer were determined using polymerase chain reaction,Western blotting,and immunohistochemical staining and their relationship with breast cancer histological grade,lymph node metastasis and ultrasonographic features was analyzed.Results&Conclusion:The expression level of RyR2 in invasive breast cancer tissues was significantly lower than that in adjacent breast cancer tissues and normal breast tissues(all P<0.01),and its low expression in invasive breast cancer tissues was associated with high histological grade of breast cancer,estrogen receptor negativity,microcalcification and lymph node metastasis(all P<0.05).Combination of ultrasonography with RyR2 detection results can provide more accurate interpretation results for the diagnosis of invasive breast cancer.
