摘 要:研究以5 dpf斑马鱼为对象,采用22 mg/mL葡萄糖(GLU)单独或与0.02和0.04 mM四氧嘧啶(ALX)溶液联合孵育24 h探索建立了基于生理和病理生理的糖代谢异常斑马鱼模型。结果显示,相比于0 mg/mL GLU组,22 mg/mL GLU单独或与0.02和0.04 mM ALX联合处理组斑马鱼GLU值分别增加了6.80倍、7.52倍和9.15倍,差异均显著(p<0.05)。采用临床常用糖尿病治疗药物阿卡波糖进行评价,发现采用22 mg/mL GLU与0.02 mM ALX联合诱导可以成功并有效建立糖尿病斑马鱼模型。基于生理和病理生理(糖尿病)的斑马鱼模型评价中,相比22 mg/mL GLU组,250、500和750μg/mL青钱柳叶水提物可以显著降低22 mg/mL GLU诱导的斑马鱼GLU值,分别降低了41.67%、48.33%和60.97%,差异均显著(p<0.05);相比22 mg/mL GLU与0.02 mM ALX联用模型组,250μg/mL青钱柳叶水提物治疗可以显著降低糖尿病斑马鱼GLU值,降低了29.97%(p<0.05),另对体重有一定的改善作用。基于斑马鱼模型的结果表明,青钱柳叶水提物具有明显的降糖效应,其在辅助降血糖功能食品饮料开发上具有广阔的应用前景。In this study,5 dpf zebrafish were chosen,and the zebrafish models with glucose metabolism disorders based on physiology and pathophysiology were evaluated and established via the usage of 22 mg/mL glucose(GLU)alone or in combination with 0.02 and 0.04 mM alloxan(ALX)for 24 h incubation.The results showed that compared with 0 mg/mL GLU group,22 mg/mL GLU alone or combined with 0.02 and 0.04 mM ALX treatments significantly increased the GLU value by 6.80,7.52 and 9.15 times,respectively(p<0.05).Acarbose is a clinical drug of treating diabetes mellitus.The zebrafish as a diabetic model,induced by 22 mg/mL combined with 0.02 mM ALX,were successfully and effectively established by the verification of hypoglycemic effect of acarbose.The results showed that,compared with 22 mg/mL GLU group,the treatments of 250,500 and 750μg/mL of the aqueous extract of Cyclocarya paliurus leaves significantly decreased the level of GLU by 41.67%,48.33% and 60.97%(p<0.05),respectively.Compared with the model group treated with 22 mg/mL and 0.02 mM ALX,250μg/mL of the aqueous extract of Cyclocarya paliurus leaves significantly reduced the level of GLU by 29.97%(p<0.05),and it had a slightly improvement effect on body weight.The research showed that the aqueous extract of Cyclocarya paliurus leaves had a significant hypoglycemic effect based on zebrafish as a model with glucose metabolism disorders.It have a broad prospect in the development of auxiliary hypoglycemic functional food and beverage.
