摘 要:沙棘是一种绿色生态资源,在我国北方地区被广泛用于水土保持和沙漠绿化。沙棘叶(Hippophae rhamnoides Linn.leaf,HRLL)是一种量大的可再生资源,已纳作普通食品原料管理。研究以HRLL为对象,采用5 dpf斑马鱼生物模型,在研究斑马鱼对HRLL水提物耐受性的基础上,开展了HRLL的降糖作用评价。结果发现斑马鱼对高浓度的HRLL具有不耐受性,孵育24 h时HRLL的LC_(50)为2479.41μg/mL,95%可信范围为2081.20~3034.93μg/mL。按照10%~25%LC_(50)剂量浓度推荐并略作调整,选用200、400和800μg/mL HRLL开展基于正常生理的和病理生理的糖代谢异常斑马鱼模型的降糖作用评价。在10 mg/mL葡萄糖诱导斑马鱼模型中,200、400和800μg/mL HRLL处理组斑马鱼葡萄糖值分别降低了33.06%(p<0.05)、37.54%(p<0.05)和54.29%(p<0.05);在10 mg/mL葡萄糖和0.025 mM四氧嘧啶诱导的糖尿病斑马鱼模型中,200、400和800μg/mL HRLL处理组斑马鱼葡萄糖值分别降低了42.22%(p<0.05)、64.47%(p<0.05)和59.49%(p<0.05)。结果表明,基于斑马鱼生物模型评价,HRLL具有明确的降糖作用,其在辅助降血糖代茶饮开发上具有广阔的应用前景。Hippophae rhamnoides Linn.is a kind of green ecological resource,which is widely used in soil and water conservation and desert greening in China’s northern areas.Hippophae rhamnoides Linn.leaf(HRLL)is a renewable resource,which has been permitted and used as a common food raw material in China.In this study,HRLL was used as the object,and a 5 dpf zebrafish biological model was used to evaluate the hypoglycemic effect of HRLL on the basis of studying the tolerance of zebrafish to HRLL water extracts.The results showed that the zebrafish were intolerant to high concentrations of HRLL water solution.The LC_(50) value was 2479.41μg/mL,and the 95% confidence range was 2081.20~3034.93μg/mL at 24 h incubation.According to 10%~25% LC_(50) with slight adjustments,200,400 and 800μg/mL HRLL were used to evaluate the hypoglycemic effect of zebrafish models with abnormal glucose metabolism based on normal physiology and pathophysiology.In 10 mg/mL glucose-induced zebrafish model,the glucose values in 200,400 and 800μg/mL HRLL treatments decreased by 33.06%(p<0.05),37.54%(p<0.05)and 54.29%(p<0.05),respectively.In the groups of diabetic zebrafish induced by 10 mg/mL glucose and 0.025 mM ALX,the glucose values in 200,400 and 800μg/mL HRLL treatments decreased by 42.22%(p<0.05),64.47%(p<0.05)and 59.49%(p<0.05),respectively.The results showed that HRLL had definite hypoglycemic effects based on the evaluation of zebrafish biological model.It has broad application prospects in the development of functional food and beverage to assist hypoglycemic.
