摘 要:目的:探讨新型光敏剂黑色二氧化钛b-TP-700介导的光动力学反应对前列腺癌细胞株PC-3的体外杀伤作用。方法:利用固相原位热还原法成功制备b-TP-700。用31.25、62.5、125、250、500、1000μg/ml b-TP-700处理PC-3细胞12、24、48 h,采用CCK-8法检测细胞活性,荧光共聚焦显微镜观察PC-3细胞形态,判断b-TP-700对PC-3细胞的毒性。采用31.25、62.5、125、250、500μg/ml b-TP-700处理PC-3细胞后,激光(1 W/cm,808 nm)照射细胞1、3、5、7 min,采用CCK-8法检测细胞活性,荧光共聚焦显微镜检测250μg/ml b-TP-700培养的PC-3细胞经激光照射5 min后细胞活性氧的产生情况,以未经激光照射的细胞为对照组。结果:不同浓度b-TP-700处理PC-3细胞相同时间时,细胞活性差异均无统计学意义(F值分别为1.26、0.39、0.37,均P>0.05)。使用荧光显微镜观察1000μg/ml b-TP-700作用PC-3细胞24 h后,细胞未出现明显形态学改变,也未见明显的死细胞。随着激光激发时间的延长,相同浓度b-TP-700作用的PC-3细胞活性下降。在激光激发时间相同时,随着b-TP-700浓度的增加,PC-3细胞的活性下降(均P<0.05)。在激光照射下,250μg/ml b-TP-700作用的PC-3细胞中可以检测到明显绿色荧光,对照组细胞中几乎观察不到绿色荧光。结论:新型光敏剂b-TP-700具有良好的活性氧生成能力,其介导的光动力学作用体外对前列腺癌PC-3细胞具有明显的杀伤作用,有望成为前列腺癌治疗的新方法。Objective To investigate the killing effects in vitro of a new photosensitizer black titanium dioxide b-TP-700-mediated photodynamics on prostate cancer cell line PC-3 cells.Methods The b-TP-700 was successfully prepared by using solid phase in situ thermal reduction method.PC-3 cells were treated with 31.25,62.5,125,250,500,1000μg/ml b-TP-700 for 12,24,48 h.The cell activity was detected by using CCK-8 method,and the morphology of PC-3 cells was observed by using confocal fluorescence microscope,the toxicity of b-TP-700 to PC-3 cells was also judged.PC-3 cells were treated with different concentrations of b-TP-700(31.25,62.5,125,250,500μg/ml)and irradiated with laser(1 W/cm,808 nm)for 1,3,5,7 min.The cell activity was detected by using CCK-8 method.The production of reactive oxygen species in PC-3 cells cultured in 250μg/ml b-TP-700 after laser irradiation for 5 min was detected by using confocal fluorescence microscopy.The non-laser irradiation cells were selected as the control group.Results There were no statistically significant differences in cell activity of PC-3 cells treated with different concentrations of b-TP-700 for the same time(F value was 1.26,0.39 and 0.37,respectively;all P>0.05).PC-3 cells were treated with 1000μg/ml b-TP-700 for 24 h,no obvious morphological changes and no obvious dead cells were observed under fluorescence microscope.When PC-3 cells were treated with the same concentration of b-TP-700,the activity of PC-3 cells was decreased with the extension of laser excitation time.Under the same laser excitation time,the activity of PC-3 cells was decreased with the increase of b-TP-700 concentration(all P<0.05).Under laser irradiation,obvious green fluorescence could be detected in the PC-3 cells treated with 250μg/ml b-TP-700,while almost no green fluorescence could be observed in the control group.Conclusions A new photosensitizer b-TP-700 has the good reactive oxygen generation ability.b-TP-700-mediated photodynamics show obvious killing effects on prostate cancer PC-3 cells in v
