摘  要:四环素类药物是一类广谱抗菌药物,在畜牧业、渔业中得到广泛应用。该药物的不合理使用易导致环境及动物源性食品中药物残留超标,从而威胁公共卫生安全。液相色谱串联质谱法是检测畜禽产品中四环素类药物残留的主要方法之一。本实验采用液相色谱-质谱联用技术(Liquid Chromatography-Mass Spectrometry/Mass Spectrometry,LC-MS/MS)检测虾粉中四环素含量,获得空白虾以及不同浓度四环素标准物质的色谱图。根据四环素浓度和峰面积的线性关系建立了标准曲线,并求出回归方程,线性良好,R^(2)=0.998 914。对比空白虾样和添加四环素的虾样,差异明显,对回收率和精密度分析,回收率为85%~103%,相对标准偏差为5.8%~9.4%。能力验证结果显示17-F701虾粉样品Z值为+1.1;17-T228虾粉样品Z值为+0.8,均达到满意结果。实验结果表明,优化后的实验条件符合要求,同时可以保证实验数据准确、可信,适用于虾中四环素的测定。Tetracyclines are a class of broad-spectrum antibacterial drugs that are widely used in animal husbandry and fishery. The unreasonable use of this drug can easily lead to excessive drug residues in the environment and animal-derived food, thus threatening public health security. Liquid chromatography-tandem mass spectrometry is one of the main methods for the detection of tetracycline residues in livestock and poultry products. In this experiment, liquid chromatography-mass spectrometry(LC-MS/MS) was used to detect the content of tetracycline in shrimp meal, and the chromatograms of blank shrimp and tetracycline standard substances with different concentrations were obtained. A standard curve was established according to the linear relationship between tetracycline concentration and peak area, and the regression equation was obtained. The linearity was good, R^(2)=0.998 914. Comparing the blank shrimp sample and the shrimp sample added with tetracycline, the difference was obvious. The recovery rate and precision analysis showed that the recovery rate was 85%~ 103%, and the relative standard deviation was 5.8%~9.4%. The proficiency testing results showed that the Z value of the 17-F701 shrimp meal sample was +1.1;the Z value of the 17-T228 shrimp meal sample was +0.8, both of which achieved satisfactory results. The experimental results show that the optimized experimental conditions meet the requirements, and the experimental data can be guaranteed to be accurate and credible, which is suitable for the determination of tetracycline in shrimp.
