摘　　要：目的 旨在阐明IQ结构域GTPase激活蛋白3(IQGAP3)在上皮性卵巢癌中的作用。方法 采用免疫组化、实时荧光定量PCR(RT-PCR)分析IQGAP3在上皮性卵巢癌细胞中的表达。通过细胞迁移实验检测IQGAP3对细胞迁移能力的影响。采用细胞侵袭实验检测IQGAP3对细胞侵袭能力的影响。通过细胞黏附检测试剂盒检测IQGAP3对细胞黏附能力的影响。采用细胞免疫荧光染色检测IQGAP3在卵巢癌细胞上的定位。结果 IQGAP3及其mRNA在上皮性卵巢癌组织中高表达。IQGAP3提高了细胞的迁移、侵袭能力,减少了细胞间黏附。IQGAP3与在上皮性卵巢癌细胞分裂中提供动力学作用的肌球蛋白在收缩环上共定位。结论 IQGAP3促进上皮性卵巢癌细胞的迁移和侵袭。Objective To clarify the role of IQ domain GTPase activator protein 3(IQGAP3) in epithelial ovarian cancer. Methods The expression of IQGAP3 in epithelial ovarian cancer was detected by immunohistochemistry and RT-PCR.The effect of IQGAP3 on cell migration was detected by cell migration experiment. The effect of IQGAP3 on cell invasion was detected by cell invasion experiment. The effect of IQGAP3 on cell adhesion was detected by cell adhesion detection kit.The localization of IQGAP3 in ovarian cancer cells was detected by cellular immunofluorescence staining. Results IQGAP3 and its mRNA highly expressed in epithelial ovarian cancer. IQGAP3 improved cell migration and invasion, and reduced intercellular adhesion. IQGAP3 is colocalization on the contractile ring with myosin, which provides a dynamic role in epithelial ovarian cancer cell division. Conclusion IQGAP3 promotes the migration and invasion of epithelial ovarian cancer cells.
