摘 要:目的探讨谷氨酸离子型受体AMPA亚基2(GRIA2)基因多态性与中国北方地区汉族人群精神分裂症易感性之间的关联。方法提取中国北方地区汉族精神分裂症患者150例(病例组)和健康志愿者160例(对照组)全血DNA,并采用iMLDR多重SNP分型技术检测GRIA2基因rs4302506、rs3813296、rs4260586和rs4441804位点的基因型。使用SPSS 23.0软件对纳入位点的数据进行统计分析,构建5种遗传模型,使用Haploview(4.2)软件进行连锁不平衡分析以及单倍型分析,并将样本按照性别、年龄分组进行等位基因、基因型频率计算。结果GRIA2基因的rs4302506、rs3813296、rs4260586和rs4441804位点的等位基因、基因型和单倍型频率在病例组和对照组间的分布差异无统计学意义(P>0.05)。按照性别、年龄分组,rs4260586位点的等位基因频率和基因型频率在男性、女性组间的差异具有统计学意义(P<0.05),携带有T等位基因和TT基因型的男性患精神分裂症的风险会升高。结论GRIA2基因的rs4260586位点与中国北方汉族人群男性精神分裂症的易感性相关。Objective To investigate the association between the polymorphisms of glutamate ionotropic receptor AMPA type subunit 2(GRIA2) gene and the susceptibility of schizophrenia(SCZ) in Han population in Northern China.Methods Whole blood DNA samples were extracted from 150 Northern Han patients of schizophrenic(case group) and 160 healthy Han volunteers(control group). The genotypes of rs4302506, rs3813296, rs4260586 and rs4441804 in GRIA2 gene were detected by the improved Multiple Ligase Detection Reaction(iMLDR). SPSS 23.0 software was used for statistical analysis of the data of the included four polymorphisms, and 5 genetic models were constructed. Linkage disequilibrium analysis and haplotype analysis were performed by Haploview(4.2) software, and the samples were grouped according to gender and age to calculate the allele and genotype frequencies. Results There were no significant difference in allele, genotype and haplotype frequencies of rs4302506, rs3813296, rs4260586 and rs4441804 in GRIA2 gene between the case and control group(P>0.05). The allele frequency of rs4260586 was significantly different between male and female groups(P<0.05), and the men carrying the T allele had an increasing risk of schizophrenia;the genotype frequency of rs4260586 was significantly different between male and female groups(P<0.05), and the men carrying the TT genotype had an increasing risk of schizophrenia. Conclusion The rs4260586 polymorphism of GRIA2 gene was associated with the susceptibility of male schizophrenia in northern Chinese Han population.
