摘 要:目的研究海带多糖对大鼠海绵体神经损伤后海绵体纤维化及勃起功能影响机制。方法选取健康ACL雄性大鼠45只,数字法随机分为H组(健康组大鼠)、M组(海绵体神经损伤模型组大鼠)及K组(海绵体神经损伤大鼠给予海带多糖干预组大鼠)。生理仪测定大鼠勃起功能;Masson染色法观察阴茎海绵体组织纤维化;ELISA测大鼠血清中H_(2)S水平;免疫组化法测大鼠阴茎组织中NOS阳性表达;免疫印记检测大鼠阴茎组织中GPX、NT-3蛋白表达。结果H组大鼠ICP/MAP比值高于M组(t=12.130,P<0.001),M组ICP/MAP比值低于K组(t=13.392,P<0.001);H组大鼠海绵体组织中胶原纤维/肌纤维比例低于M组(t=8.668,P<0.001),M组大鼠胶原纤维/肌纤维比例高于K组(t=6.193,P<0.001);H组大鼠血清H_(2)S水平在不同时间点均高于M组(P<0.05),M组大鼠血清H_(2)S水平在不同时间点均低于K组(P<0.05);在不同时间点H组大鼠阴茎组织中NOS阳性表达均高于M组(P<0.05),M组大鼠阴茎组织中NOS阳性表达均低于K组(P<0.05);不同时间点,H组大鼠阴茎组织中GPX、NT-3蛋白均低于M组(P<0.05),M组大鼠阴茎组织中GPX、NT-3蛋白水平均高于K组(P<0.05)。结论海带多糖通过提高H_(2)S、NOS水平,降低GPX、NT-3表达,促进海绵体神经损伤修复,降低海绵体组织纤维化水平,改善大鼠勃起功能障碍。Objective To study the mechanism of kelp polysaccharide on cavernous fibrosis and erectile function after cavernous nerve injury in rats. Methods A total of 45 healthy ACL male rats were selected. By digital method, they were randomly divided into group H(healthy group), group M(cavernosal nerve injury model group) and group K(cavernosal nerve injury rats treated with kelp polysaccharide intervention group). The erectile function of rats was measured by physiological apparatus. Masson staining was used to observe the tissue fibrosis of penile cavernous body. The levels of H_(2)S in serum of rats were measured by ELISA. Immunohistochemical method was used to detect the positive expression of NOS in rat penile tissue. The expression of GPX and NT-3 protein in rat penile tissue was detected by immunoimprinting. Results The ratio of ICP/MAP in group H was higher than that in group M(t=12.130, P<0.001), and the ratio of ICP/MAP in group M was lower than that in group K(t=13.392, P<0.001). The proportion of collagen fiber/muscle fiber in the cavernosal tissue of rats in group H was lower than that in group M(t=8.668, P<0.001), and the proportion of collagen fiber/muscle fiber in group M was higher than that in group K(t=6.193, P<0.001). The serum H_(2)S level of group H was higher than that of group M at different time points(P<0.05), and the serum H_(2)S level of group M was lower than that of group K at different time points(P<0.05).The positive expression of NOS in penile tissue of rats in group H was higher than that in group M at different time points(P<0.05), and the positive expression of NOS in penile tissue of rats in group M was lower than that in group K(P<0.05). At different time points, the protein levels of GPX and NT-3 in penile tissue of rats in group H were lower than those in group M(P<0.05), and the protein levels of GPX and NT-3 in penile tissue of rats in group M were higher than those in group K(P<0.05).
