摘  要:[目的]探讨配对样同源转录因子2(PITX2)在胃癌组织及胃癌细胞中的表达,以及对细胞增殖、迁移和侵袭的影响。[方法]收集50例胃癌患者胃癌组织及癌旁组织,于液氮中封存,通过RT-qPCR和Western-blot实验检测胃癌组织及癌旁组织PITX2表达情况,并分析其与患者临床病理因素之间的关系。通过RT-qPCR检测正常胃上皮细胞GES-1、胃癌细胞系AGS、SGC-7901、MGC-803中PITX2的表达水平,选取表达量最高的MGC-803细胞为研究对象,分为siRNA-PITX2组、siRNA-NC组以及对照组,通过CCK8检测各组细胞增殖情况,划痕实验检测各组细胞迁移情况,Transwell检测各组细胞侵袭情况,Western-blot检测各组细胞Ki-67、CyclinD1、MMP-2、MMP-9蛋白表达水平。[结果]与癌旁组织和胃上皮细胞比,PITX2在胃癌组织和胃癌细胞系中表达水平显著升高(P<0.05),PITX2高表达与患者年龄、性别、肿瘤大小均无关,与肿瘤转移和TNM分期有关(P<0.05);干扰PITX2表达后,MGC-803细胞增殖能力、迁移和侵袭能力均显著下降,Ki-67、CyclinD1、MMP-2、MMP-9蛋白表达水平下降(均P<0.05)。[结论]PITX2在胃癌组织和胃癌细胞中高表达,干扰PITX2表达能够抑制胃癌细胞MGC-803增殖、迁移和侵袭。[Objective]To investigate expression of PITX2 and its effects on proliferation and invasion in gastric cancer tissues and cell lines.[Methods]Tumor tissues and adjacent tissues of 50 patients diagnosed with gastric cancer between 2015 and 2018 were collected and sealed in liquid nitrogen.The expression level of PITX2 in gastric cancer tissues and adjacent tissues was detected by RT-qPCR and western-blot, and the relationship between PITX2 and clinicopathological factors was analyzed.The expression levels of PITX2 in normal gastric mucosa cell line GES-1,gastric cancer cell lines AGS,SGC-7901,and MGC-803 were detected by RT-qPCR.MGC-803 cell lines with highest level in PITX2 were divided into siRNA-PITX2 group, siRNA-NC group, and control group.Cell proliferation in each group was detected by CCK8,and cell migration was observed by scratch test.Cell invasion in each group was detected by transwell, and Ki-67,CyclinD1,MMP-2,and MMP-9 protein expression levels in each group were detected by Western-blot.[Results]Compared with adjacent tissues and gastric epithelial cells, the expression levels of PITX2 were significantly increased in gastric cancer tissues and gastric cancer cell lines(P<0.05),and the high expression of PITX2 was not related to age, gender, or tumor size, but was related to tumor metastasis and TNM stage(P<0.05);after interfering with PITX2 expression, the proliferation, migration, and invasion of MGC-803 cells were significantly decreased, and the protein expression levels of Ki-67,CyclinD1,MMP-2,and MMP-9 were decreased(P<0.05).[Conclusion]PITX2 is highly expressed in gastric cancer tissues and gastric cancer cells, and interference with PITX2 expression could inhibit the proliferation, migration and invasion of gastric cancer cell.
