摘  要:目的:以网络药理学为基础,对归芍地黄汤治疗慢性再生障碍性贫血的分子作用机制进行探索挖掘。方法:通过中药系统药理学数据库与分析平台(Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform,TCMSP)查找药物的有效成分,进行药代动力参数筛选,通过Swiss Target Prediction数据库找出有效化合物的作用靶点;在Gene Cards、OMIM、TTD、Dis Ge NET、Mala Cards数据库查找慢性再生障碍性贫血的主要靶点;通过String数据库分析蛋白质之间的相互作用,并构建蛋白质相互作用网络。使用Metascape数据库分析药物参与的生物过程及发挥作用的细胞通路,采用Cytoscape软件构建药物化合物-靶点-通路网络,进行可视化分析。最后使用Autodock Vina进行分子对接验证。结果:归芍地黄汤治疗慢性再生障碍性贫血的核心化学成分为猪苓酸C、槲皮素、羟基芫花素、山柰酚及茯苓新酸C等,核心靶点为丝裂原活化蛋白激酶1(Mitogen-Activated Protein Kinase 1,MAPK1)、肿瘤抑制p53蛋白(Tumor Protein P53,TP53)、HRas原癌基因(HRAS)、NRAS原癌基因(NRAS)及淀粉样蛋白β前体蛋白(APP)等。主要的作用机制为MAPK途径的激活促进细胞增殖及显著的抗炎作用。结论:本研究探寻了归芍地黄汤治疗慢性再生障碍性贫血多种成分、多靶点及多通路的作用机制,为临床上该药物的使用及开发提供理论依据。Objective:The action mechanism of the Guishao Dihuang decoction(归芍地黄汤)on chronic aplastic anemia was explored based on network pharmacology.Methods:The active ingredients of medicines were searched in TCMSP,and ADME screening was conducted.The effective compound targets were searched in the SwissTargetPrediction database.The main targets of chronic aplastic anemia were searched in GeneCards,OMIM,TTD,DisGeNET and MalaCards databases.The interaction between proteins was analyzed by String database,and a PPI network was constructed.The biological process in which the medicine participated and cellular pathways in action were analyzed by Metascape database.A‘medicine compound-target-pathway’network was constructed by Cytoscape software for visual analysis.Finally,Autodock Vina was used for molecular docking verification.Results:The core chemical components of the Guishao Dihuang decoction on chronic aplastic anemia were polyporic acid C,quercetin,hydroxychrysanthemum,kaempferol,poriacin C and so on.The core targets were MAPK1,TP53,HRAS,NRAS,APP and so on.The main action mechanism is the activation of the MAPK pathway to promote cell proliferation and significant anti-inflammatory effects.Conclusion:This study explored the multi-component,multi-target and multi-pathway action mechanism of the Guishao Dihuang decoction on chronic aplastic anemia,and provided a theoretical basis for its clinical application and development.
