摘 要:目的:观察染料木素磺酸钠(Genisteinsodium sulfonate,GSS)是否可通过调节Notch1信号减少慢性脑缺血大鼠皮层神经元的凋亡。方法:采用改良型双侧颈总动脉结扎法(2-VO)制备慢性脑缺血大鼠模型,运用尼氏染色方法观察大鼠皮层神经元损伤情况,并以Real time-PCR和Western Blot方法检测大鼠皮层组织中Caspase-3、Bcl-2、Bax等凋亡相关分子mRNA和/蛋白表达,同时检测Notch1的蛋白表达。结果:经GSS治疗后,慢性脑缺血大鼠皮层神经元尼氏小体数量明显增多,并且下调了大鼠大脑皮层组织中的促凋亡分子Caspase-3和Bax的mRNA及蛋白表达水平,上调抗凋亡分子Bcl-2的mRNA及蛋白表达水平,GSS还可有效激活2-VO大鼠皮层组织中Notch1的蛋白表达。结论:GSS可能通过激活Notch1信号减轻慢性脑缺血所诱导的神经元凋亡性损伤。Objective:To observe whether genisteinsodium sulfonate(GSS)can regulate Notch1 signal to reduce the apoptosis of cortical neurons in chronic cerebral ischemia rats.Methods:The modified bilateral common carotid artery ligation(2-VO)method was used to prepare the rat model of chronic cerebral ischemia.Nissner’s staining method was used to observe the injury of cortical neurons in rats.The mRNA and protein expression levels of Caspase-3,Bcl-2,Bax were detected by Real time-PCR and Western Blot,and the activity of Notch1 was also detected by Western Blot.Results:After GSS treatment,Nissner’s bodies of cortical neurons in chronic cerebral ischemia rats were significantly increased.In addition,GSS down-regulated the mRNA and protein expression levels of pro-apoptotic molecules Caspase-3and Bax and up-regulated the mRNA and protein expression of the anti-apoptotic molecules Bcl-2 in cerebral cortex tissue of rats.GSS could also effectively activate the protein expression level of Notch1 in the cortical tissues of 2-VO rats.Conclusion:GSS may reduce neuron apoptosis induced by chronic cerebral ischemia by activating the Notch1 signal.
