摘 要:为探究蒙古黄芪种子贮藏过程中活力和生理生化指标的变化,揭示其老化机理、筛选适宜贮藏的含水量和温度,以不同含水量(3.7%,6.6%,9.8%)的蒙古黄芪种子为试材,于不同条件(室内、短期库、中期库、长期库)下贮藏12个月,测定种子发芽率和生理生化指标。结果表明,不同贮藏条件和不同含水量的种子发芽率均不断降低,在这4种条件下贮藏12个月后,含水量为9.8%的种子发芽率分别为27.7%、39.3%、42.3%、47.3%;含水量为6.6%的种子发芽率分别为32.3%、42.3%、44.0%、53.3%;含水量为3.7%的种子发芽率分别为35%、44.7%、49.7%、62.0%;贮藏过程中种子活性氧含量升高,SOD活性升高,APX活性不断下降,有毒物质MDA逐渐积累,可溶性蛋白和可溶性糖含量下降;种子含水量和贮藏温度协同影响种子活力,低含水量且低温条件更有利于保持种子活力,大幅减小种子贮藏过程中发芽率的下降幅度。因此,蒙古黄芪种子适宜贮藏条件为:含水量3.7%、温度-18℃。To explore the vigor and physiological and biochemical changes of Astragalus mongolicus seed during storage process,to reveal the mechanism of aging and screen suitable water content and temperature for storage,the seeds of A.mongholicus with different water content(3.7%,6.6%and 9.8%)were stored for 12 months in different conditions(indoor,short-term storage,medium-term storage and long-term storage),and the germination rate and physiological and biochemical indexes of the seeds were determined.The results showed that the germination rate of seeds under different storage conditions and different water content were decreased continuously.After 12 months storage under the four conditions,the germination rate of seeds with 9.8%water content were 27.7%,39.3%,42.3%and 47.3%,respectively.The germination rate of seeds with 6.6%water content were 32.3%,42.3%,44.0%and 53.3%,respectively.The germination rates of seeds with 3.7%water content were 35%,44.7%,49.7%and 62.0%,respectively.During the storage process,the content of reactive oxygen increased and the activity of SOD increased while the activity of APX decreased.In addition,the toxic substance MDA accumulated gradually,and the content of soluble protein and soluble sugar decreased.Seed water content and storage temperature had a synergistic effect on seed vigor.Low water content and low temperature were more conducive to maintaining seed vigor,which greatly reduced the decrease of seed germination rate during seed storage.Therefore,the suitable storage conditions of A.mongolicus seeds were 3.7%water content and-18℃temperature.
