摘 要:目的分析探讨甲硫氨酸亚砜还原酶A(MsrA)在肾透明细胞癌和癌旁组织中的表达及其临床意义。方法利用免疫组织化学染色方法检测并观察比较MsrA在5对肾透明细胞癌及其癌旁组织中的蛋白表达差异,并结合癌症基因组图谱(TCGA)数据库,分析MsrA的表达量与肾透明细胞癌患者临床数据特征之间的关系,应用Kaplan-Meier和Cox回归研究其表达水平与临床预后之间的关系,利用列线图构建一预测肾透明细胞癌患者的生存预后模型。结果MsrA主要表达在肾小管上皮细胞质中且在癌旁组织的蛋白表达量更高。TCGA数据库结果显示,MsrA在总样本和配对样本中肾透明细胞癌组织的mRNA表达量明显低于癌旁组织,且MsrA在女性、低级别、低临床分期中的表达量更高。生存分析显示,MsrA高表达组患者生存率更高;Cox比例风险回归分析表明MsrA表达、年龄、分级和临床分期是影响肾透明细胞癌患者预后的独立生存因子。结论MsrA表达是肾透明细胞癌患者预后的独立预测因子,可成为其潜在诊断和治疗的生物标志物。Objective To investigate the expression and clinical significance of methionine sulfoxide reductase A(MsrA) in renal clear cell carcinoma and adjacent tissues. Methods Immunohistochemical staining was used to detect and compare the protein expression of MsrA in 5 pairs of renal clear cell carcinoma and their adjacent tissues. The relationship between the expression level of MsrA and the characteristics of renal clear cell carcinoma patients was analyzed using The Cancer Genome Atlas(TCGA) database. Kaplan-meier and Cox regression analyses were used to investigate the relationship between the expression level of MsrA and clinical prognosis in renal clear cell carcinoma. The nomogram was utilized to construct a prognosis model for the renal clear cell carcinoma patients. Results MsrA was mainly expressed in the cell cytoplasm of renal tubular epithelium and its level was higher in the adjacent tissues. The mRNA level of MsrA in renal clear cell carcinoma tissues was significantly lower than that in adjacent tissues in total and paired samples based on the TCGA database. The expression of MsrA was higher in female, low-grade and low clinical stages. Survival analysis showed that patients with high expression of MsrA had favorable survival outcomes. Cox proportional hazard regression analysis showed that MsrA expression, age, grade, and clinical stage were independent predictors in renal clear cell carcinoma patients. Conclusion MsrA expression is an independent predictor and could be a potential biomarker for diagnosis and treatment in renal clear cell carcinoma patients.
