摘 要:目的探究微小RNA(microRNA,miR)-103b靶向Flotillin-2(FLOT2)调节肾癌细胞生物学行为的作用机制。方法通过双荧光素酶报告验证miR-130b与FLOT2的靶向关系。人肾癌细胞分为4组:对照组、miR-103b组、FLOT2组和miR-103b+FLOT2组。通过转染miR-103b类似物和/或FLOT2质粒来提高miR-103b和/或FLOT2的水平。检测各组FLOT2蛋白表达水平,以及细胞增殖、迁移和侵袭。结果 miR-103b与FLOT2在肾癌细胞中靶向结合(P<0.05);miR-103b组的FLOT2蛋白、增殖、迁移和侵袭水平显著低于对照组(P<0.05);FLOT2组的FLOT2蛋白、增殖、迁移和侵袭水平显著高于对照组(P<0.05);miR-103b+FLOT2组的FLOT2蛋白、增殖、迁移和侵袭水平显著高于miR-103b组且显著低于FLOT2组(P<0.05)。结论 miR-103b可能通过靶向FLOT2抑制肾癌的进展。Objective To explore the mechanism of microRNA(miR)-103 b targeting Flotillin-2(FLOT2)to regulate the biological behavior of renal cancer cells. Methods The target relationship between miR-130 b and FLOT2 was verified by dual luciferase report. Human kidney cancer cells were divided into 4 groups:control group,miR-103 b group,FLOT2 group and miR-103 b+FLOT2 group. The level of miR-103 b and/or FLOT2 were increased by transfection of miR-103 b analog and/or FLOT2 plasmid. The expression level of FLOT2 protein in each group,as well as cell proliferation,migration and invasion were tested. Results The miR-103 b and FLOT2 targeting bound in renal cancer cells(P<0.05). The levels of FLOT2 protein,proliferation,migration and invasion in the miR-103 b group were significantly lower than those in the control group(P<0.05). The levels of FLOT2 protein,proliferation,migration and invasion in the FLOT2 group were significantly higher than those in the control group(P<0.05). The levels of FLOT2 protein,proliferation,migration and invasion in the miR-103 b+FLOT2 group were significantly higher than those in the miR-103 b group and significantly lower than those in the FLOT2 group(P<0.05). Conclusion The miR-103 b can inhibit the progression of kidney cancer by targeting FLOT2.
