摘  要:目的研究铁棒锤水提物对大鼠的急性毒性作用,为临床用药安全和后续实验提供参考。方法清洁级Wistar雄性大鼠56只,随机分为7组:对照组(生理盐水)、铁棒锤水提物(15、30、60、120、240和480 mg/kg)剂量组,连续灌胃给药处理14 d,观察大鼠14 d内所产生的毒性反应,并记录大鼠给药前后体质量和进食变化情况;第15天处死大鼠,通过测定血液常规指标以及血清生化指标丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)和肌酐(CRE),计算心脏、肝、肾和脑脏器系数,测定各组织中丙二醛(MDA)含量,苏木素—伊红染色观察各组织脏器病理学变化,评价其急性毒性作用。结果与对照组相比,铁棒锤水提物剂量组大鼠体质量、饲料消耗量、食物利用率及各组织脏器系数差异均无统计学意义(P>0.05),铁棒锤水提物480 mg/kg剂量组血清ALT、AST、CRE含量均高于对照组(P<0.05),但均在大鼠正常生理值范围内,其余给药组血清生化指标无明显变化。铁棒锤水提物剂量组大鼠血液常规指标、各组织MDA含量差异无统计学意义,组织病理切片亦未见异常现象。结论大鼠经口给予铁棒锤水提物的最大给药量为480 mg/kg,为成人推荐日用最大剂量的576倍,安全性良好,表明铁棒锤经水煮炮制的水提物具有一定的安全范围,为铁棒锤的临床应用提供一定的可能性和安全性。Objective To study the acute toxic effect of water extract from Aconitum pendulum Busch in rats,in order to provide reference for the safety of clinical medication and subsequent experiments.Methods Totally56 clean grade Wistar rats were randomly divided into 7 groups:the control group(normal saline),water extract from Aconitum pendulum Busch(15,30,60,120,240 and 480 mg/kg)dose group by way of irrigation to continuous processing rats 14 days.The toxicity reaction of the rats were observed within 14 days,and each animal medication changes before and after the body quality and eating were recorded;rats were executed in15 days’time;the blood routine index,serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),creatinine(CRE)and conventional hematological indexes were measured.The organ coefficients of heart,liver,kidney and brain were calculated to determine the content of malondialdehyde(MDA)in each tissue,the pathological changes of each tissue were observed by hematoxylin-eosin staining and the acute toxic effects were evaluated.Results Compared with the control group,there were no statistically significant differences in the body mass,feed consumption and food utilization rate and organ coefficients of each tissue in the water extract dose group from Aconitum pendulum Busch(P>0.05).The contents of in serum of ALT,AST and CRE in the water extract dose group of 480 mg/kg from Aconitum pendulum Busch were all higher than those in the control group(P<0.05),but all were within the normal physiological value range of rats,with the rest of the drug group of biochemical indicators having no obvious change.There were no statistically significant differences in conventional hematological indicators and MDA content in each tissue,and no abnormalities were found in the histopathological sections.Conclusion The maximum dose of water extract from Aconitum pendulum Busch was 480 mg/kg,which was 576 times of the daily dose of adults,indicating that the water extract from Aconitum pendulum Busch processed by water
