摘 要:以铜绿假单胞菌(Pseudomonas aeruginosa)FC-69为出发菌株,以排油圈直径和生物表面活性剂产量为考核指标,采用紫外线和亚硝基胍复合诱变,筛选到一株生物表面活性剂高产菌株FC-69-52,并对其遗传稳定性和特性进行了研究。结果表明,高产菌株FC-69-52的排油圈直径达到7.12 cm、生物表面活性剂产量达到42.86 g·L^(-1),分别较出发菌株FC-69提高了14.84%和19.06%;高产菌株FC-69-52发酵48 h,生物表面活性剂产量达到42.21 g·L^(-1),乳化值达到65.68%,发酵液表面张力降至27.05 mN·m^(-1),对油水界面的亲和力更强,可使乳化相在96 h内保持稳定,可作为微生物发酵法产生物表面活性剂的优良高产菌株。Using Pseudomonas aeruginosa FC-69 as a starting strain,and the diameter of oil drain ring and the production of biosurfactant as evaluation indexes,we screened a high-yield biosurfactant-producing strain FC-69-52 by UV and nitrosoguanidine compound mutagenesis,and studied its genetic stability and characteristics.The results show that,the diameter of oil drain ring of the high-yield strain FC-69-52 is 7.12 cm,and the production of biosurfactant is 42.86 g·L^(-1),which is 14.84%and 19.06%higher than that of the starting strain FC-69,respectively.After 48 h fermentation of the high-yield strain FC-69-52,the production of biosurfactant reaches 42.21 g·L^(-1),the emulsification value reaches 65.68%,and the surface tension of the fermentation broth decreases to 27.05 mN·m^(-1),respectively.The high-yield strain FC-69-52 has stronger affinity for the oil-water interface and can keep the emulsified phase stable within 96 h.It can be used as an excellent high-yield strain for the production of biosurfactants by microbial fermentation.
