番鸭细小病毒NS和VP基因的克隆及序列分析

摘  要:为进一步了解番鸭细小病毒(MDPV)基因的遗传变异特征,本试验采用分段PCR扩增法获得了2个MDPV安徽分离株(AH1901和AH1902)基因组NS和VP全长基因,并与GenBank数据库中的20个水禽细小病毒基因组序列进行了比对。序列分析显示,克隆的MDPV基因均包括完整的NS和VP基因,分别为1884 bp和2199 bp,VP基因核苷酸序列变异程度相对较高,而NS基因则较为保守;进化分析显示,安徽分离株AH1901和AH1902位于同一分支,二者与国内分离的MDPV毒株ZJ-JH-2013基因遗传距离最近,可能来源于同一毒株,与MDPV疫苗株P1及国外分离毒株FM基因遗传距离则较远;安徽分离株AH1901和AH1902的NS基因序列同源性为99.8%,VP基因序列一致。两者的NS、VP基因序列与分离株ZJ-JH-2013的同源性最高,与重组型毒株SAAS-SHNH及安徽分离株AH1401同源性也在99.5%以上;与疫苗株P1、国外分离株FM同源性则较低,其中VP基因仅为88.9%、89.3%。本试验结果表明近年国内的MDPV分离株同源性均较高,但仍存在一定程度的基因变异,增加了诊断与防控该病的难度。To investigate the mutational status of Muscovy duck parvovirus(MDPV)genes,this study utilized PCR to amplify the two NS and VP full-length genes of MDPV isolated in Anhui,and compared their sequences with those of 20 waterfowl parvoviruses in GenBank.The results showed that the amplified fragments contained full-length NS and VP genes of 1884bp and 2199 bp,respectively.The VP sequence was somewhat variable,while the NS sequence was relatively conservative.Evolutionary analysis revealed that Anhui isolates AH1901 and AH1902 were clustered in the same branch,had a close genetic relationship with domestic strain ZJ-JH-2013,suggesting their similar origin,and displayed genetic distance from MDPV vaccine strain P1 and Hungary strain FM.Furthermore,Anhui isolates AH1901 and AH1902 shared 99.8%similarity in the NS gene,and were identical in the VP gene.Additionally,Anhui isolates AH1901 and AH1902 demonstrated highest homology with isolate ZJ-JH-2013,at least 99.5%homology with recombinant strain SAAS-SHNH and Anhui isolates AH1401,and only 88.9%and 89.3%with strain P1 and FM in the VP gene.Thus,despite sharing high homology,MDPV still shows a certain degree of variation,which may add complexity to its diagnosis and prevention.
