摘  要:目的研究槐耳清膏对三阴性乳腺癌细胞增殖和迁移的作用及其可能的机制。方法采用CCK-8法及小鼠移植瘤模型检测槐耳清膏对乳腺癌细胞增殖的影响,细胞划痕实验检测槐耳浸膏对乳腺癌细胞迁移的影响。Western blot检测细胞蛋白表达的水平。结果CCK-8结果表明,采用4.16、6.48、10.37 mg/mL的槐耳清膏分别处理人三阴性乳腺癌MDA-MB-231、MDA-MB-453细胞及小鼠三阴性乳腺癌4T1细胞24h后,细胞活力被显著抑制(P<0.05),且呈浓度和时间依赖性;划痕实验显示,24 h IC_(50)浓度的槐耳浸膏处理人三阴性乳腺癌MDA-MB-231细胞及小鼠三阴性乳腺癌4T1细胞24 h可显著抑制细胞划痕的愈合(P<0.05)。Western blot法显示IC_(50)浓度槐耳清膏作用24 h后,细胞中N-Cad-herin的水平为对照组的79%、68%、61%;E-Cadherin水平显著高于对照组(P<0.05)。HIPPO通路效应蛋白YAP蛋白水平为对照组的51%和64%,存在显著差异(P<0.0001),其Ser109点位的磷酸化蛋白表达水平提高176%。经槐耳浸膏灌胃的三阴性乳腺癌荷瘤小鼠的肿瘤体积明显小于对照组(P<0.05)。结论槐耳清膏可抑制三阴性乳腺癌细胞的增殖和迁移,其机制与HIPPO通路被抑制相关。Objective To study the effect of Huaier aqueous extract on proliferation and migration of the triple-negative breast cancer cells and its possible mechanism. Methods CCK-8 method and mouse xenograft model were used to detect the effect of Huaier aqueous extract on cell proliferation, and the cell scratch test was used to detect the effect of Huaier aqueous extract on cell migration. Western blot was used to detect the level of cell protein expression. Results CCK-8 assays showed that treated the MDA-MB-231, MDA-MB-453 cells of human triple-negative breast cancer and the 4 T1 cells of mice triple-negative breast cancer with 4.16, 6.48 and 10.37 mg/mL Huaier aqueous extract for 24 hours significantly inhibited cell viability in a concentration and time-dependent manner(P<0.05). The healing of cell scratches was significantly inhibited after the MDA-MB-231 and the 4 T1 cells were respectively treated with Huaier aqueous extract at IC_(50) for 24 h(P<0.05). Western blot showed that after treatment of Huaier aqueous extract for 24 h, the levels of N-Cadherin in the cells were 79%, 68%, and 61% of those in the control group;The levels of E-Cadherin were significantly higher than those in the control group, and the differences were all significant. The levels of effector protein YAP in the HIPPO pathway were 51% and 64% of those in the control group, with significant difference(P<0.0001). The triple-negative breast cancer tumor-bearing mice treated by Huaier aqueous extract were significantly smaller than those of the control group, and there was a significant difference(P<0.05). Conclusion Huaier aqueous extract can inhibit the proliferation and migration of triple-negative breast cancer cells, and its mechanism is related to the inhibition of the HIPPO pathway.
